<h3>Abstract</h3>
Published in 2022, the <a href="https://gcms.labrulez.com/library?search=flow-modulated+GCxGC-FID">flow-modulated GCxGC-FID</a> ASTM Standard Test Method D8396 for <a href="https://gcms.labrulez.com/library?search=Group-Type+Quantitation">Group-Type Quantitation of Hydrocarbons</a> in Hydrocarbon Liquids demonstrates that GCxGC is gaining acceptance as a routine technique in the petroleum industry. 
With this acceptance comes the expectation for easy-to-understand, user-friendly interfaces and robust hardware that reliably provides full transfer of analytes throughout the separation space for accurate quantitation of chemical species that correlate to bulk physical properties of petroleum products. Preliminary results from testing both gravimetric standards and a variety of reference aviation and diesel fuels evaluate a variety of method parameters crucial to providing proper transfer of analytes from the primary to secondary column when using flow modulation. 
Clear workflows that simplify GCxGC method development for reverse fill-flush flow modulation are optimized for easier adoption in routine laboratories. Additional support in characterization of samples is provided using a prototype GCxGC-TOFMS/FID instrument which is equipped with a novel splitter design that ensures a consistent split ratio between the MS and FID detectors to avoid erroneous area percent determinations as well as a novel peak alignment algorithm that simplifies identification of FID peaks using corresponding MS spectra to assist in drawing regions for more accurate group-type quantitation of chemical classes.

Method development for petroleum analysis using GC×GC-MS/FID (Sebastiano Panto, MDCW 2023)

Los Angeles, California —LECO Corporation has partnered with California State University, Los Angeles, on the development of its new LECO Complex Chemical Compositional Analysis Lab. The opening of the lab on September 19th marked a significant step towards advancing research and education in complex chemical analysis not only at the university but throughout the West Coast.
The new C3AL, as it is referred to, is located in the renovated laboratory of Rosser Hall at CSULA. Here, students, colleagues, and partners will study the composition and structure of complex chemical samples under the direction of Dr. Petr Vozka, professor in the Department of Chemistry and Biochemistry.

As part of its contribution to the new laboratory, LECO provided state-of-the-art time-of-flight mass spectrometry equipment. The Pegasus BT brings <a href="https://gcms.cz/products/36">GC</a>, <a href="https://gcms.cz/products/37">GCxGC</a>, and TOFMS capabilities to the laboratory and allows for deep investigations to a variety of complex samples and interesting questions. Having access to this technology provides the students at the C3AL an opportunity to collaborate with and learn from industry leaders currently using these same tools to solve real-world problems.
LECO, based in St. Joseph, Michigan, plans to utilize the lab for future training and product demonstrations on the West Coast as well.

<h5>Proteomics:</h5>
We are modern well equipped laboratory with the technical background able to perform most of the currently used <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/untargeted-proteomics.24?t=s">untargeted</a> and <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/targeted-proteomics.25?t=s">targeted</a> proteomic measurements. We provide full proteomics service, including protein isolation from the supplied sample to evaluation and consultation of the measured data. Since 2015, we handle approximately 300 orders per year (roughly 3000 samples). Results based on measurements from our laboratory were used in dozens of publications. <h5>Metabolomics:</h5>
As a service, we provide <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/targeted-metabolomics.26?t=s">targeted</a> and <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/untargeted-metabolomics.30?t=s">untargeted</a> analysis of small molecules. Thanks to the combination of equipment for LC-MS and GCxGC-MS, it is possible to measure a wide range of small molecules. We use several types of C18 and HILIC stationary phase columns on LC. For GC, we have various columns for different types of analytes. Individual measuremnts must always be consulted in advance, an individual approach and method must be chosen for each group of molecules. <h4>Untargeted proteomics</h4>
<h5>Up to 8000 identified proteins in one injection.</h5>
A basic type of proteomic experiment. Complete lysate is digested with trypsin. The resulting peptides are desalted and separated on a reverse phase column. Due to the low amount of proteins and the high complexity of the sample, liquid chromatography is currently most often used in nanoflow - hundreds of nanoliters per minute. Liquid chromatography is directly connected to the electrospray of the Orbitrap Fusion or Orbitrap Ascend mass spectrometer. The mass spectrometer measures alternately in two modes: MS¹, where peptide masses are measured in the Orbitrap with high resolution and accuracy, and MS², in which peptides selected from the previous MS¹ scan are fragmented. These are measured either again in the Orbitrap or in the ion trap. The ion trap provides lower accuracy and resolution than the Orbitrap, but is faster and more sensitive.

<h4>Targeted proteomics</h4>
<h5>Accurate quantification of selected proteins.</h5>
Method aims to validate the results from an untargeted approach. It is performed on a triple quadrupole in SRM mode or using the MRM method on an Orbitrap. It is an an alternative approach to common techniques such as western blot. The advantage of this approach is the possibility to analyze several proteins (dozens) at once during one injection. This eliminates the problem of finding suitable antibodies.

The targeted proteomics service is always project specific and we offer it after prior meeting with the customer.

Parallel analysis of peptides labeled with heavy amino acids is essential. These serve as an internal standards.
The amount of proteins that can be targeted in one injection using a triple quadrupole, is strongly dependent on the concentration of the target proteins in the sample. The lower the expression of the target proteins, the less it is possible to analyze them in one injection due to the need for higher loading times. Information of fragmentation properties of targeted peptides is also important. This is usually obtained from an untargeted experiment, which should precede the targeted proteomic analysis. If the protein of interest was not detected during an untargeted experiment, then its targeted analysis is very difficult. However, in some cases fragmentation information can be obtained from publicly available databases.

<h4>Untargeted metabolomics</h4>
The goal of untargeted metabolomics is to cover the widest possible spectrum of metabolites present in the sample, their identification, and quantification. The development of this approach was made possible by the recent advances in instrumentation and computing. Our laboratory equipment enables non-targeted analyses by liquid and gas chromatography.

We offer untargeted analyses of metabolites in various biological matrices and basic statistical evaluation of the data (Figure 2, 3, 4).

Our newest service is stable isotope tracing (Figure 1).

HPLC-MS/MS (Figure 2): Analyzes are performed on Thermo Scientific equipment on an Ultimate 3000 liquid chromatograph coupled to an Orbitrap Fusion mass detector. The measured data are evaluated by the Compound Discoverer software.
GCxGC-MS (Figure 3): Analyzes are performed on a two-dimensional comprehensive chromatograph connected to a Pegasus 4D mass detector. We evaluate the data in ChromaTOF v4.5 software.
The liquid chromatography method covers non-volatile substances, generally polar, soluble in water, and polar organic solvents (methanol, acetonitrile...). The gas chromatography method is more suitable for analyzing stable volatile non-polar substances up to a weight of approx. 1000 Da. The range of measured substances can be increased for both methods eg., by chemical modifications (derivatization) of the analyzed molecules, which adjust their polarity and volatility (for GC) and/or stability (for LC). We choose a suitable technique or a combination of several techniques based on prior consultation with the customer.
The untargeted analysis results in a list of substances found in the samples, a proposal of probable structures, and an indication of the quantity of individual compounds. We also provide a basic statistical evaluation focusing on looking for differences between the tested sample groups (Figure 4).

<h4>Targeted metabolomics</h4>
Targeted metabolomics includes analyses of selected metabolites, which are then detected selectively and with greater sensitivity than applying an untargeted approach. It is also possible to quantify substances both relatively and absolutely. In the case of absolute quantification, it is necessary to supply a suitable labeled analog of the determined substance.

The attached list contains substances with the analysis of which we have experience. We adapt each method to the type of experiment and matrix. We can develop a method for any substance or group of substances capable of being detected by liquid and gas chromatography techniques (both connected to a mass detector or a UV detector in liquid chromatography). In the case of the development of a new method, it is necessary to supply the standards of these substances.

The mass spectrometer enables improved sample throughput, versatility and ease-of-use — producing high quality data with simplicity. More samples can be analyzed at lower concentrations, enabling faster, more sensitive analysis, and macromolecule experiments, that were not possible before, can now be achieved. The new Auto-Ready ion source for calibration maximizes convenience, allowing the user to spend more time on the results, and less time on instrument set-up.

Mass spectrometer combining Quadrupole, Ion trap and Orbitrap. The quadrupole is used as a mass filter for the selection of precursors, after which the instrument branches and the ions can travel either to the Orbitrap - a very accurate and high-resolution mass analyzer that uses the Fourier transform to determine their mass, or to the collision cell, where they can be fragmented. From there, the path leads either back to the Orbitrap or further to the linear ion trap. This serves as the second mass analyzer in the device. It is less accurate and low-resolution, but provides greater sensitivity and the possibility of ion fragmentation. Fragmentation products can be isolated in the trap and fragmented again. Introducing BIOCEV OMICS Mass Spectrometry Core Facility

When a police dog is tasked with finding a missing person, which odors will guide its search? And which combination of chemicals forms those odors?<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/articles?search=Katelynn+Perrault+Uptmor">Katelynn Perrault Uptmor</a>, assistant professor of chemistry at William &amp; Mary, answers questions like these in her research into the chemical breakdown of substances involved in forensics (such as decomposing remains or fingerprint residue) and food (including coffee and kombucha). Now, she has a new, state-of-the-art instrument to elevate her work.In June 2024, Perrault Uptmor won a national contest and was awarded an advanced piece of lab equipment: a brand-new <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=24&amp;search=BTX">Leco Pegasus BTX 4D</a> with Paradigm Shift Flow Modular from Leco Corp., one of the leaders of separation science tools.<figure class="image"><img style="aspect-ratio:1620/1080;" src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/William_and_Mary_Expanding_Research_Chemistry_professor_Katelynn_Perrault_Uptmor_center_stands_with_research_students_at_her_lab_in_the_Integrated_Science_Center_Photos_courtesy_of_Katelynn_Perrault_Uptmor_910df5bd16.jpg" alt="William &amp; Mary: Expanding Research: Chemistry professor Katelynn Perrault Uptmor (center) stands with research students at her lab in the Integrated Science Center. It will increase the capacity of her lab’s productivity, provide additional research opportunities for students and enable her to solve more complex research questions.“I'm not the chemist who sits at a bench mixing chemicals together to make something new. I'm the chemist who tries to find out what something is made of,” says Perrault Uptmor.She arrived at William &amp; Mary in August 2023 after an academic world tour: After growing up in Toronto, she moved to Australia to complete a Ph.D., traveled to Belgium for postdoctoral training and accepted her first faculty position at Chaminade University of Honolulu in Hawaii. At W&amp;M, she is excited about having increased resources, time for research and students who are eager to embark on chemistry research.The new equipment from Leco will further energize her work, which utilizes a process called <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=2&amp;mainauthorItems=24">comprehensive two-dimensional gas chromatography (GC×GC)</a> to identify the components of chemical samples.<img style="aspect-ratio:667/500;" src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/William_and_Mary_The_new_Leco_equipment_arrived_at_Perrault_Uptmor_s_lab_in_the_summer_of_2024_1a94f66c0f.jpg" alt="William &amp; Mary: The new Leco equipment arrived at Perrault Uptmor's lab in the summer of 2024" srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_William_and_Mary_The_new_Leco_equipment_arrived_at_Perrault_Uptmor_s_lab_in_the_summer_of_2024_1a94f66c0f.jpg 208w," sizes="100vw" width="667" height="500">“Based on the lab’s needs, the importance of the work being done and the genuine passion for GC×GC, we believe she is the most deserving of this state-of-the-art equipment,” says John Hayes, separation science product manager at Leco.&nbsp;The process of gas chromatography allows Perrault Uptmor to gain a more comprehensive separation of all the chemicals in a sample and therefore acquire more specific data. The Leco instrument is further advanced in this process than alternative separation science tools, with superior sensitivity and sustainability.&nbsp;Emma Macturk, a graduate student in chemistry who studies the composition of fingerprint residue in Perrault Uptmor’s lab, says that the gift will significantly improve the lab’s ability to conduct separation research: “With more sophisticated instrumentation, we can ask and answer more complex questions.”GC×GC instruments contain tubes (called capillaries) through which samples travel as they are separated into their different components. The capillaries in many conventional GC×GC instruments are not long enough to separate the hundreds of compounds in Perrault Uptmor’s complex samples. But the Leco GC×GC instrument contains two capillaries connected via an interface, enabling better detection and sensitivity. &nbsp;The new instrument also provides a more sustainable process of research, a feature that both Perrault Uptmor and Macturk emphasize. The improved sustainability stems from the instrument’s modulator, which enables the process of separation. Other instruments use temperature to control separation — increasing heat to speed up the process and cooling to slow it down — and requires a constant supply of liquid cryogens. But the new Leco instrument uses valves to stop and release flow, a much greener approach. The old approach costs money and time and resources, Perrault Uptmor says, while the new system avoids those constraints.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/William_and_Mary_Perrault_Uptmor_started_teaching_and_researching_in_the_chemistry_department_at_W_and_M_in_August_2023_e148c57701.jpg" alt="William &amp; Mary: Perrault Uptmor started teaching and researching in the chemistry department at W&amp;M in August 2023." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_William_and_Mary_Perrault_Uptmor_started_teaching_and_researching_in_the_chemistry_department_at_W_and_M_in_August_2023_e148c57701.jpg 202w," sizes="100vw" width="647" height="500">The mechanical details of the new equipment are exciting; the consequent increased capacity for her lab and the impact on her students’ research is even more so, she says.“This is a huge capacity build that allows us to essentially run double the number of projects, which means more undergraduate students get training on it,” says Perrault Uptmor.Professor of Chemistry J.C. Poutsma, a fellow analytical chemist, is similarly enthusiastic about the impact of the equipment on student education and training: “We are delighted that Leco chose Kate to receive the new mass spectrometer. The acquisition of this state-of-the-art instrument will allow our undergraduate chemistry majors to experience cutting-edge mass spectrometry techniques in both classroom and independent research settings.”Perrault Uptmor currently has seven undergraduates and one graduate student working in her lab. Students often begin as volunteers before signing up to do research for credit and working in the lab over the summer “My goal is for them to be fully responsible for their own project, take ownership of that project and get an authentic research experience,” she says.While students take the lead on individual projects, there is still frequent collaboration in the lab. And students have one-on-one instruction with Perrault Uptmor as well, often working through data analysis strategies and software development.Kira Fisher ’26, a chemistry major, works in Perrault Uptmor’s lab developing an easy workflow for method development with the GC×GC process. Fisher arrived at chemistry research after an initial interest in forensic science. I feel comfortable analyzing data on my own and doing daily lab tasks by myself — something I was not comfortable doing before joining this lab,” Fisher says.Sarah Foster ’27, another student in Perrault Uptmor’s lab, got an early start in the lab in the fall of her first year at W&amp;M. Foster is working on two research projects: analyzing volatile organic compounds (VOCs) present in fresh versus fermented poi (a traditional Polynesian food) and analyzing VOCs in different brands of kombucha. For the kombucha project, she is using the new Leco instrument.“It is really amazing to have the opportunity to work with new instrumentation like the Leco instruments we have in our lab,” Foster says. “At first, I was nervous to work with such fancy equipment, but Dr. Kate really helped me feel comfortable around everything. It is a real confidence boost to be trusted to run the instruments and take care of regular lab maintenance.”This past summer, Foster accepted a summer fellowship with the Food and Drug Administration through the Oak Ridge Institute for Science and Education, where she performed bioanalytical chemistry research.Experience with lab work and this type of instrumentation will be beneficial for W&amp;M chemistry students’ career options.“A large majority of companies want students that are trained in analytical chemistry. And a lot of schools are actually minimizing their analytical departments,” says Perrault Uptmor.With the addition of the Leco instrument, Perrault Uptmor’s lab will distinguish the W&amp;M chemistry department from those at other universities and ensure that students can develop highly marketable job skills. “From a research and teaching perspective,” she says, “this is really exciting.”

January 2026</blockquote><h3>Abstract</h3>Ultrafine particles (UFP) have diameters below 100 nm and a high surface area. As they may enter the lung deeply, UFP pose a potential risk to human health. Efforts in sampling and analysing UFP are challenged by their low mass. Thus, to gain insights into their chemical composition, efficient sampling strategies and highly sensitive instrumentation are needed.Ambient air UFP was sampled in two field campaigns in March and September 2023 in the city of Augsburg, Germany. Samples were collected on filters for 24 h and analysed via direct thermal desorption coupled to comprehensive two-dimensional gas chromatography time-of-flight mass spectrometry (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=2-13-3-8">DTD-GC×GC-TOFMS</a>, <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=25&amp;search=LECO+BT4D">LECO BT4D</a>). Compound classes of known adverse health effects, e.g. polycyclic aromatic hydrocarbons (PAHs) including alkylated, oxygenated 4 and 5-ring PAHs and parent-4, 5 and 6-Ring PAHs were identified. Ten PAHs, including Benz[a]pyrene (BaP) - as one of the most ubiquitous and cancerogenic PAHs - were subsequently quantified. We found BaP concentrations of 6-14 pg/m3 and 4-38 pg/m3 in March and September, respectively. Moreover, the classification approach demonstrated relatively higher abundances of parent-4,5 and 6-ring PAHs as well as alkylated and oxygenated 4- and 5-ring PAHs during two days of high BaP concentration.This study gives novel insights into the composition of ambient air UFP and highlights the importance of applying sophisticated analytical techniques such as GC×GC-MS to gain a comprehensive understanding of complex sample composition.This project was financed by the Bavarian Ministry of the Environment and Consumer Protection.

January 2026</blockquote><h3>Abstract</h3>Over the past decade, metabolomics has gained significant importance in the life sciences, offering insights into metabolic pathways involved in various pathological processes. As this technology becomes crucial in research, the need to enhance and standardize quality assurance (QA) and quality control (QC) practices in untargeted metabolomics has intensified. Despite efforts to develop QA/QC systems, such as those of the Metabolomics Quality Assurance and Quality Control Consortium (mQACC), challenges remain, particularly in terms of documentation and standardization, which are essential for advancing non- targeted metabolomics.In our research, we focused on establishing a robust QA/QC system for untargeted analysis using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=2-13-3&amp;mainauthorItems=24">GC×GC-TOFMS</a> systems. We analyzed the Century Mix (CM), a QC solution mix developed by the FDA comprising 100 diverse compounds, to assess analytical performance. The method effectively monitored compounds with varying volatility and chemical functions, demonstrating good distribution and separation across the 2D chromatogram during a 60-minute run on different instruments with varying column sets.To ensure the reliability of our analytical processes, we implemented a sophisticated QA/QC system featuring control charts that tracked retention time and response (area values) for all compounds over six months. Our goal is to extend this systematic approach to all four GC×GC systems in our laboratory, across various sample types and studies, to establish a standardized and robust QA/QC framework using the CM, thereby enhancing the precision and reproducibility of untargeted metabolomics research.
