LECO / MDCW: BOOSTING NON-TARGETED ANALYSIS WITH COMPREHENSIVE TWO-DIMENSIONAL GAS CHROMATOGRAPHY AND HIGH-RESOLUTION MASS SPECTROMETRY</h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/FL_18_Sebastiano_b4cf417f82.pdf">Full PDF for download</a></li></ul>The presentation by Sebastiano Panto (LECO EATC Berlin) focuses on demonstrating how comprehensive two-dimensional gas chromatography coupled with high-resolution time-of-flight mass spectrometry (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=2-13-3&amp;mainauthorItems=24">GC×GC-HR-TOFMS</a>) significantly enhances non-targeted and targeted screening workflows, particularly in environmental analysis. Using liver extracts from Glaucous gulls (Larus hyperboreus) from the Svalbard region as a case study, the work aims to reveal contamination patterns and improve compound identification confidence.Sample preparation included homogenization, lipid removal, and multi-step cleanup involving GPC/SEC chromatography and Florisil fractionation, yielding separate non-polar and polar fractions for analysis (page 4). Analytical separations were performed on an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890+GC">Agilent 8890 GC</a> equipped with a dual-column GC×GC setup (Rxi-5SilMS and Rxi-17SilMS), cryogenic modulation, and Helium carrier gas. Detection used the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=24&amp;search=Pegasus+HRT%2B4D+HR-TOFMS">LECO Pegasus® HRT+4D HR-TOFMS</a> with multiple ionization modes—EI, PCI, and ECNI—enabling simultaneous acquisition of structural, isotopic, and exact-mass information (page 5).The results highlight the advantages of GC×GC-HR-TOFMS in non-target screening: improved separation patterns, higher library match scores, exact-mass confirmation, and enhanced structural elucidation. Electron ionization (EI) provided clean spectra with high match scores for classic pollutants (e.g., PCBs, DDE), while electron capture negative ionization (ECNI) greatly improved selectivity and sensitivity for halogenated contaminants such as toxaphenes and brominated flame retardants, enabling confident identification even when EI spectra were ambiguous (pages 6–13). The instrument’s high mass accuracy and isotopic fidelity are shown to be essential for distinguishing closely related halogenated compounds.The study concludes that combining GC×GC separation power with HR-TOFMS accurate-mass detection and complementary ionization modes offers a robust discovery platform for environmental exposomics. This analytical workflow supports more confident annotation of persistent organic pollutants (POPs) and emerging contaminants in complex biological matrices, ultimately helping assess ecological risks in sensitive Arctic ecosystems where these avian species reside (page 15).<h3>3. Shimadzu: High-Sensitivity Gas Analysis Using the GCMS-QP2050 and GI-30 Auto Gas Injector</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00953_en_7e1a865aae.pdf">Full PDF for download</a></li></ul>Gas analysis is conducted across a wide range of fields, including resources, energy, and the environment. Typical targets include inorganic gases such as H2, CO, and CO2, and light hydrocarbon gases starting with CH4, with analyses primarily performed using GC. In Application News No. 01- 00858, examples of gas sample analysis using TCD and BID installed on the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Brevis+GC-2050">Brevis GC-2050</a> were presented.&nbsp;However, qualitative analysis by GC requires standard substances, making the identification of unknown impurities in gas samples difficult. Furthermore, in inorganic gas analysis, complete chromatographic separation of peaks can be challenging, and when GC fails to fully separate them, both qualitative and quantitative analysis become difficult.&nbsp;In contrast, gas analysis using GC-MS enables qualitative identification of unknown components without standard substances through mass spectral library searches. Quantitation in GC-MS is performed using mass chromatograms, allowing accurate measurements even when peaks overlap.&nbsp;In this Application News, high-sensitivity analysis of inorganic gases and light hydrocarbon gases was performed using the compact yet high-performance <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-QP2050">GCMS-QP2050</a>. Gas samples were introduced using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GI-30">GI-30 Auto Gas Injector</a>, enabling automated, continuous analysis and delivering higher reproducibility than manual sample injection.<h4>Instrument Configuration and Analytical Conditions&nbsp;</h4>A 10 ppm standard mixed gas sample in a cylinder was analyzed using the GCMS-QP2050. Gas sample injection into the GC-MS was performed with the GI-30 Auto Gas Injector (P/N: S221- 89755-41). The instrument configuration is shown in Fig. 1. The analytical conditions used are summarized in Table 1. Under typical ionization settings (ionization voltage 70 V and emission current 60 µA), ionization of the helium carrier gas generates a significant signal at m/z 2, making detection of H2 (monitor ion: m/z 2) difficult. Therefore, for analysis in SIM mode, the ionization voltage was set to 20 V and the emission current to 20 µA to suppress He ionization.<h4>Automated Continuous Analysis Using the GI-30&nbsp;</h4>The GI-30 Auto Gas Injector is a sample pretreatment device that automatically introduces a fixed volume of gas into the GC. It is used by connecting either a gas cylinder or a sample bag. When the GI-30 is connected to a gas cylinder, automated, continuous analysis of gassamples becomes possible.&nbsp;The GI-30 is equipped with an optional valve purge mechanism. Purging the internal valve with gas reduces the ingress of ambient air. This is effective for analyzing components in ambient air, such as N2 and O2, as well as for measuring tracelevel components.&nbsp;Table 2 shows the peak-area repeatability (%RSD) obtained when a 10 ppm standard mixed gas was analyzed automatically and continuously in SIM mode. As an example, an overlay of the CH4 SIM chromatograms is shown in Fig. 3. Because the GI-30 meters a fixed volume of gas into the GC via a sample loop, excellent reproducibility was achieved for all compounds.<h4>Conclusion&nbsp;</h4>Using the GCMS-QP2050, high-sensitivity analysis of inorganic gases, such as H2, and light hydrocarbons was achieved. Gas sample injection was performed with the GI-30 Auto Gas Injector, enabling automated, continuous analysis and delivering higher reproducibility compared with manual sample injection. The compact, high-performance GCMS-QP2050, together with the GI-30, enables automation and high-precision gas analysis, and supports greater efficiency in GC-MS-based gas analysis.<h3>4. Thermo Fisher Scientific: Trace analysis of epichlorohydrin in drinking water using GC-MS coupled with purge and trap</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_003894_gc_single_quad_isq_7610_environmental_an003894_na_en_70123385ae.pdf">Full PDF for download</a></li></ul>Epichlorohydrin (ECH) is a versatile starting material in the production of drugs and polymers and is also used as an insect fumigant and solvent for organic synthesis reactions. ECH-based polymer pipes are commonly used in the production of drinking water due to their durability and resistance to corrosion. However, ECH is known for its high reactivity and toxicity, which poses significant health risks if it contaminates drinking water. Exposure to ECH can cause respiratory issues, skin irritation, and has been classified as a probable human carcinogen.Due to these risks, many countries have imposed strict limits on the amount of ECH allowed in drinking water. Recently, Europe set a minimum detection limit (MDL) of 30 parts per trillion (ppt) for ECH in drinking water.2 Whereas typically required detection limits for most compounds mandated for analysis in Europe can be achieved using methods based on static headspace, the stringent MDL required for ECH requires preconcentration, for example using purge and trap (P&amp;T) technology.3,4 This technology involves purging the water sample with an inert gas to release volatile organic compounds (VOCs), which are then trapped and concentrated for analysis, ensuring reliable detection at very low concentrations. In the United States, the analysis of VOCs in drinking water is mandated by the Environmental Protection Agency (EPA). The EPA requires the use of P&amp;T technology for drinking water analysis to ensure that even trace amounts of harmful compounds like ECH are detected and managed appropriately.&nbsp;The following evaluation describes the use of the ISQ 7610 Single Quadrupole MS system coupled with the Thermo Scientific™ TRACE™ 1610 GC with the Thermo Scientific™ HeSaver-H2 Safer™ split/splitless injector and Teledyne LABS Tekmar Lumin P&amp;T concentrator combined with the AQUATek LVA autosampler for the analysis of ECH in drinking water.<h4>Experimental</h4><h5>GC-MS parameters&nbsp;</h5>A <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/486">TRACE 1610 GC</a> was coupled to the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/484">ISQ 7610 MS</a> equipped with the Thermo Scientific™ NeverVent ™ vacuum probe interlock (VPI) and an ExtractaBrite ion source. A <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/1773">Thermo Scientific™ TraceGOLD™ TG-VMS column</a>, 30 m × 0.25 mm, 1.4 µm film (P/N 26080-3320) was used for compound separation. The HeSaver-H2 Safer SSL injector was utilized to reduce the carrier gas consumption by decoupling the gas used for the chromatographic separation from the gas used to pressurize the inlet and maintain split and purge flows. The critical separations were maintained with a run time of under 15 minutes.For this analysis, the ISQ 7610 MS was operated in Selected Ion Monitoring (SIM) mode for increased selectivity, as required for this application. Extended method parameters for the ISQ 7610 MS are shown in Table 2.<h5>Instrument control and data processing&nbsp;</h5>Data was acquired, processed, and reported using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/76">Thermo Scientific™ Chromeleon™ Chromatography Data System (CDS)</a>. This software can control both the GC-MS system and the Tekmar Lumin P&amp;T with the AQUATek LVA. This allows a single software to be utilized for the full workflow, simplifying the instrument operation. This application note is available for download via the Thermo Scientific™ AppsLab Library, which contains all the parameters needed to acquire, process, and report the analytical data for analysis of ECH.5<h4>Conclusion&nbsp;</h4>This study demonstrates the capability of the Tekmar Lumin P&amp;T with the AQUATek LVA system connected to the ISQ 7610 Single Quadrupole GC-MS to detect and quantify low-level ECH in drinking water samples, in compliance with EPA requirements.&nbsp;<ul><li>Utilizing the Tekmar Lumin P&amp;T’s ability to purge with nitrogen, along with using the HeSaver-H₂Safer SSL injector, nearly four times less helium was consumed during the analysis without sacrificing system performance.&nbsp;</li><li>The linearity of the calibration curve from 30 ppt to 5,000 ppt passed method requirements.</li><li>The application proved robust during an extended study with 20 samples of a 1,000 ppt ECH standard injected over a series of 124 injections, obtaining 4.4% precision and 118% accuracy of the recovery.</li></ul>

Durian (Durio zibethinus), known as the "king of fruits" and widely exported across Southeast Asia, China, and beyond, presents unique analytical challenges due to its sulfur-rich compounds, high fat content, and complex volatile matrix. Comprehensive pesticide analysis in such matrices demands highly sensitive, selective, and robust analytical solutions. The European Union Reference Laboratories (EURL) SANTE/11312/2021 guidelines set rigorous performance criteria for pesticide residue analysis, including strict requirements for method accuracy, precision, and sensitivity.1 Laboratories need to achieve recoveries between 70% to 120%, precision (RSD) below 20%, and quantification limits at or below regulatory maximum residue limits (MRLs), even in complex food matrices such as durian. This application note demonstrates a high-performance workflow for the analysis of 197 pesticides in durian using an Agilent PAL3 Autosampler combined with an Agilent 7000E Triple Quadrupole GC/MSD system. The PAL3 Autosampler provides high-throughput, precise sample handling, while the 7000E GC/MSD offers exceptional sensitivity and reproducibility for trace-level pesticide quantification. Optimized sample preparation and instrumental conditions ensure reliable performance, meeting and exceeding SANTE criteria for method validation.<h4>Experimental</h4><h5>PAL 3 RTC and 7000E GC/TQ parameters&nbsp;</h5>The analysis parameters are shown in Table 1. The target and ISTD compound MRM parameters are listed in Appendix&nbsp;1. The optimized workflow, including sample extraction with an Agilent Bond Elut QuEChERS EN kit followed by cleanup with the Highly Pigmented dSPE kit, provided excellent recovery and precision. All target pesticides showed analysis accuracy between 80% and 120% and RSD values below 10% at a spiked level of 10 ppb. The results confirm the method's suitability for routine high-throughput pesticide quantification in complex matrices such as durian. The use of matrix-matched calibration and matrix‑optimized MRMs further ensured accurate and reproducible quantitation, reinforcing the method's applicability for regulatory and food safety testing.<h3>2. LECO / MDCW: Applying statistical data processing tools for GC×GC differentiation of alternative aviation fuels</h3><ul><li>Presentation</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/FL_18_Sebastiano_b4cf417f82.pdf">Full PDF for download</a></li></ul>The presentation introduces LECO’s long-standing role in separation science and its evolution from elemental analyzers to advanced mass spectrometry platforms. A key emphasis is placed on LECO’s innovations in time-of-flight mass spectrometry (TOFMS), including the Pegasus series and the new <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=24&amp;search=Pegasus+BTX">Pegasus BTX</a>, a benchtop TOFMS offering high sensitivity, rugged detector technology, fast spectral acquisition (1–500 spectra/s), and broad mass range (10–1500 m/z). This technological foundation supports comprehensive GC×GC-MS workflows essential for analyzing chemically complex matrices such as aviation fuels.A major focus of the talk is on software tools for non-targeted data processing. LECO’s software ecosystem—<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=24&amp;search=ChromaTOF">ChromaTOF</a>, ChromaTOF Tile, and <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=24&amp;search=ChromaTOF+Sync">ChromaTOF Sync</a>—enables detailed deconvolution, quantitation, high-resolution spectral analysis, and especially statistical comparison of GC×GC datasets. ChromaTOF Tile uses a chromatographic “tiling” strategy to link datasets and detect differences between sample groups using three statistical metrics: Fisher Ratio, Coefficient of Variation (CoV), and Fold-Change. Each metric serves a different experimental purpose, from comparing traditional vs. synthetic jet fuels (Fisher Ratio) to exploring variability among multiple alternative fuel processes (CoV), or identifying additive components in two-sample comparisons (Fold-Change).The presentation demonstrates how raw GC×GC-MS data are curated into feature tables, statistically evaluated, and visualized in PCA plots, bar charts, and chromatographic overlays. For example, Fisher Ratio comparison highlights statistically significant chemical differences between conventional Jet A and synthetic fuels, while CoV analysis reveals clustering patterns among diverse synthetic fuel production pathways such as LT-FT SPK, HT-FT SPK, HEFA, SIP, ATJ, and CHJ. These workflows enable analysts to rapidly identify biomarkers or distinguishing compositional features within highly complex hydrocarbon matrices.Ultimately, the talk demonstrates that choosing the correct statistical processing method can dramatically simplify interpretation of complex GC×GC-MS datasets. LECO’s integrated instrumentation—particularly the Pegasus BTX TOFMS—and software platforms together provide a powerful toolkit for fuel characterization, quality evaluation, and differentiation of emerging alternative aviation fuels. By combining comprehensive chromatography with robust statistical approaches, researchers can gain deeper insights into chemical variability, production signatures, and additive packages in modern aviation fuel technologies.<h3>3. Shimadzu: Analysis of Formic Acid in Acetone Using GC/MS System</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00957_en_e8b083b86e.pdf">Full PDF for download</a></li></ul>In research on artificial photosynthesis, the analysis of impurities in chemical products and raw materials has increased the demand for high-sensitivity analysis of formic acid. Previous examples of formic acid analysis using GC-BID and GC-FID (Jetanizer ) have been presented, but in this article high-sensitivity analysis of formic acid using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-QP2050">GCMS-QP2050</a> (see Fig. 1) isintroduced.<h4>Insert, Column Phosphoric Acid Treatment&nbsp;</h4>Formic acid is known to adsorb onto the injection port and column, making peak detection difficult. However, by performing phosphoric acid treatment on the glass insert and column before measurement, good peaks can be obtained. The procedure for phosphoric acid treatment of the glass insert is shown in Fig. 2, and the procedure for phosphoric acid treatment of the column isshown in Fig. 3.<h4>Conclusion&nbsp;</h4>By performing phosphoric acid treatment on the insert and column, it became possible to analyze formic acid in SIM mode down to 0.2 ppm. Furthermore, good results could be obtained for both linearity and repeatability. Additionally, by analyzing in Scan mode, identification of unknown peaks other than formic acid was also achieved.<h3>4. Many of these compounds contaminate our environment and may cause negative health effects in humans and other living beings. Analytical laboratories must monitor a variety of sample types from the environment to ensure the public are not exposed to elevated levels of VOCs. The latest version of applicable method in the United States, U.S. EPA Method 8260C, is applicable when monitoring a variety of solid waste matrices for the presence of VOCs.To perform U.S. EPA Method 8260C, all method acceptance criteria must be achieved. These criteria include calculating the mean response factor and the relative standard deviation (RSD) of the response factors for target analytes. The RSD should be &lt;20%, with minimum response factors (RF) and MDLs for a wide range of target compounds. The analytical method must produce consistent results and be reproducible from day to day, with a continuing calibration verification (CCV) analyzed every 12 hours while samples are run. As the method covers varying matrices, it is important that the performance criteria are met in all samples of interest. The following evaluation describes the use of the ISQ 7610 GCMS coupled to the Atomx XYZ P&amp;T for U.S. EPA Method 8260C.<h4>Experimental</h4><h5>Instrument control and data processing&nbsp;</h5>Data were acquired, processed, and reported using <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/76">Chromeleon CDS software</a>, version 7.2. This software can control both the GC-MS system and the Tekmar Atomx XYZ P&amp;T. This enables a single software solution to support the full workflow, simplifying the instrument operation. The optimized method used within this application note is available for download via the Thermo Scientific™ AppsLab library. AppsLab contains all the parameters needed to acquire, process, and report the analytical data for U.S. EPA Method 8260C.2<h5>GC-MS parameters&nbsp;</h5>A <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/486">TRACE 1610 GC</a> was coupled to the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/484">ISQ 7610 MS </a>equipped with the Thermo Scientific™ NeverVent™ vacuum probe interlock (VPI) and an ExtractaBrite™ ion source. Expanded method parameters for the GC-MS system are displayed in Table 3<h4>Conclusion&nbsp;</h4>The combined analytical solution with the TRACE 1610 GC coupled with the ISQ 7610 system and the Atomx XYZ P&amp;T system provides clear advantages for analytical testing laboratories that analyze environmental samples following the U.S. EPA Method 8260C requirements. The modularity of the TRACE 1610 GC as well as the ISQ 7610 VPI and ExtractaBrite ion source allows users to easily service the injection ports and to exchange ionization sources and analytical columns without venting the mass spectrometer, significantly reducing instrument downtime and minimizing sample analysis interruptions. The Atomx XYZ concentrator’s efficient trap cooling design reduces sample cycle time and allows for increased sample throughput. The moisture control system improves water vapor removal thereby reducing peak interference and increasing GC column life span.&nbsp;The experiments performed clearly demonstrate the suitability of this analytical configuration for the analysis of VOCs in various environmental samples in accordance with U.S. EPA Method 8260C with the following performance parameters as evidence:&nbsp;<ul><li>The ISQ 7610 VPI coupled with the Tekmar Atomx XYZ P&amp;T exceeds all the requirements outlined in U.S. EPA Method 8260C for analysis of VOCs in wastewater and solid waste.</li><li>Linearity was achieved with &lt;20%,&nbsp;relative standard deviation for both water and soil calibration curves for the majority of compounds.&nbsp;</li><li>The MDL and precision were assessed using n=7 replicates of a 0.2 ppb water standard and n=7 replicates of a 0.5 ppb soil standard. Calculated MDLs were &lt;0.2ppb and RSDs of calculated results were &lt;10% for most compounds in both the soil and water matrices.</li><li>System robustness was tested by continuously acquiring 240 injections of environmental samples over three days with no user intervention at all. The average %RSD of the calculated concentration was 8.30% with an average compound recovery of 90%.</li></ul>

The Ministry of Ecology and Environment issued "The 14th Five-Year Plan for Ecological and Environmental Monitoring", which specifically highlights the advancement of technologies for direct measurement of carbon emissions. It calls for organizing pilot projects in multiple key industries and enterprises to monitor greenhouse gas emissions such as carbon dioxide and methane, establishing greenhouse gas monitoring networks in key cities, and upgrading the greenhouse gas monitoring capabilities of national atmospheric background stations. As a result, conducting greenhouse gas monitoring within the environmental protection stations of provinces and cities has become a critical task.The use of GC for detecting greenhouse gases has a long history. The conventional methods used gas sampling valve for injection, TCD and FID for sequential detection of CO2 and CH4 , or combining a methanizer with an FID to detect CO2 and CH4 in one analytical channel and detecting N2O on ECD in another analytical channel.2 The analytical precision for CO2, CH4, and N2 O achieved with such configurations typically ranges from 0.2% to 0.5%, but it is difficult to obtain precision better than 0.1%, which does not meet the current requirements for greenhouse gas monitoring accuracy and precision. Moreover, the traditional methods rarely report analytical precision for SF6 at ppt concentration levels. To monitor trace levels of CO2 , CH4, N2O, and ultratrace levels of SF6 on the same GC system with high-precision results, it is necessary to optimize multiple factors included in sample injection, separation, and detection. Furthermore, the system configuration must be flexible enough to allow expansion for future analytical needs, such as incorporating trace CO analysis in air. This work demonstrates how the 8890&nbsp;GC achieves high-precision analysis of four key greenhouse gases by optimizing the injection volume, controlling injection reproducibility, and utilizing multichannel separation and high-sensitivity detectors to ensure the system runs stably in long‑term operation.<h4>Experimental</h4><h5>Instrumental&nbsp;</h5>The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=8890+GC">8890 GC</a> is configured as a three-channel/multivalve/multicolumn system, enabling independent analysis of four greenhouse gases—CH4 , CO2 , N2O, and SF6 —within 8 minutes using a single injection. The target compounds are analyzed in separate channels, reducing chromatographic peak broadening and thereby improving sensitivity and&nbsp;repeatability.&nbsp;<ul><li>For CO2 analysis, a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?manufacturerItems=1&amp;search=methanizer">methanizer</a> is used to convert CO2 to CH4 , which is then detected by FID. The system eliminates the need for dehydration through backflushing and venting operation via switching valves, preventing interference from moisture and oxygen in the air and extending the lifespan of the methanizer.&nbsp;</li><li>For N2O and SF6 analysis, the ECD with hidden anode design is used for high‑sensitivity detection and strong anti‑contaminant capability.&nbsp;</li><li>The CH4 analysis is conducted on a single column with a dedicated large injection loop, significantly enhancing detection sensitivity and repeatability, achieving a detection limit for methane as low as single-digit ppb levels.</li></ul><h4>Conclusion&nbsp;</h4>Based on its exceptional pneumatic control and valve-based flow path design, the multivalve/multicolumn Agilent 8890 GC system achieves high-sensitivity and high-precision measurements of four key greenhouse gases. This is accomplished through optimized and precise control of sample flow rate/volume, selection of premium analytical columns, and the use of highly stable and robust FID and ECD. Standard gas samples of CO2 , CH4 , N2 O, and SF6 , with concentrations equivalent to atmospheric levels, were measured continuously 10 times. The measurement precision for these gases was better than 0.05%, 0.1%, 0.1%, and 0.5%, respectively, fully meeting the high-precision monitoring requirements of environmental monitoring stations at various levels for target greenhouse gases. Compared to other monitoring solutions, this approach is based on a stable and reliable gas chromatography platform that is technically easier to master and deploy. ECH-based polymer pipes are commonly used in the production of drinking water due to their durability and resistance to corrosion. However, ECH is known for its high reactivity and toxicity, which poses significant health risks if it contaminates drinking water. Exposure to ECH can cause respiratory issues, skin irritation, and has been classified as a probable human carcinogen.1Due to these risks, many countries have imposed strict limits on the amount of ECH allowed in drinking water. Recently, Europe set a minimum detection limit (MDL) of 30 parts per trillion (ppt) for ECH in drinking water.2 Whereas typically required detection limits for most compounds mandated for analysis in Europe can be achieved using methods based on static headspace, the stringent MDL required for ECH requires preconcentration, for example using purge and trap (P&amp;T) technology.3,4 This technology involves purging the water sample with an inert gas to release volatile organic compounds (VOCs), which are then trapped and concentrated for analysis, ensuring reliable detection at very low concentrations. In the United States, the analysis of VOCs in drinking water is mandated by the Environmental Protection Agency (EPA). The EPA requires the use of P&amp;T technology for drinking water analysis to ensure that even trace amounts of harmful compounds like ECH are detected and managed appropriately.&nbsp;The following evaluation describes the use of the ISQ 7610 Single Quadrupole MS system coupled with the Thermo Scientific™ TRACE™ 1610 GC with the Thermo Scientific™ HeSaver-H2 Safer™ split/splitless injector and Teledyne LABS Tekmar Lumin P&amp;T concentrator combined with the AQUATek LVA autosampler for the analysis of ECH in drinking water.<h4>Experimental</h4><h5>GC-MS parameters&nbsp;</h5>A <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/486">TRACE 1610 GC</a> was coupled to the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/484">ISQ 7610 MS</a> equipped with the Thermo Scientific™ NeverVent ™ vacuum probe interlock (VPI) and an ExtractaBrite ion source. Identification of controlled substances relies heavily on comparison of mass spectra to commercially available spectral libraries, and increased column bleed can cause spectral interference at high temperatures. These interferences can lead to poor library matching probabilities.1-3 The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=J%26W+5Q">Agilent J&amp;W 5Q series</a> of gas chromatography columns provide the most thermally stable columns available, which helps to decrease spectral interference due to column bleed and improves data accuracy.&nbsp;Poor thermal stability will cause an increase in the slope of the baseline as the oven temperature is increased, leading to difficult peak integrations and inaccurate quantitation. Additionally, sloped or elevated baselines can lead to a decreased response and a loss of sensitivity for analytes eluting during the oven ramp or at the method’s upper temperature limits. Several commercially available 5ms type columns were compared to the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=J%26W+DB-5Q">Agilent J&amp;W DB-5Q GC column</a> to examine the impact thermal stability has on response at upper temperature limits.&nbsp;<h4>Experimental&nbsp;</h4>An <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890+GC">Agilent 8890 GC system</a> coupled with an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=5977B">Agilent 5977B mass selective detector (GC/MSD)</a> with an inert extractor ion source was used for acquisition of data and MassHunter 10.0 Quantitative and Qualitative Analysis software was used for data analysis.&nbsp;A representative toxicology mixture, the Agilent GC/MS forensic toxicology checkout mixture (part number 5190-0471) was used to monitor analytical performance. Calibration standards were prepared in methanol (MeOH) at concentrations ranging from 0.1 to 3.0&nbsp;μg/mL. An internal standard mix was prepared at 10&nbsp;μg/mL from individual stocks of methamphetamine-d3, diazepam-d5, and hydrocodone-d6 all purchased from Millipore Sigma (St.&nbsp;Louis, MO).<h4>Results and discussion&nbsp;</h4>The testing evaluated the Agilent J&amp;W DB-5Q GC column for the analysis of controlled substances by GC/MS. The increased thermal stability of the DB-5Q is seen in the decreased baseline at higher temperatures, as seen in Figure&nbsp;1, and also provides a decrease in the slope during the oven ramping program.&nbsp;An increase in the slope of the baseline will also decrease the signal-to-noise ratio (S/N) of an analyte but will also make integration more difficult and lead to less accurate quantitative results. Figure 2 demonstrates the comparison of the DB-5Q and the commercially available 5ms type G column, and the impact to chromatography during the oven ramp. The S/N for the analytes on the oven ramp slope are a factor of ten or greater for the DB-5Q in comparison to the 5ms type G column.<h4>Conclusion&nbsp;</h4>The increased thermal stability of the Agilent J&amp;W DB-5Q GC column demonstrated in this application note provides a decreased baseline at higher temperatures, which allows for an increased response for later eluting compounds at trace level concentrations. The baseline of the DB-5Q remains more consistent as the temperature is increased. This consistency allows for more accurate integration and quantitation for analytes eluting during oven ramping, in comparison to other commercially available 5ms-type columns. The robust DB-5Q column phase maintained peak shape for trace level basic compounds even when stressed with a heavy matrix and is an excellent choice for the analysis of controlled substance&nbsp;analysis.<h3>2. Its chemical structure is shown in Fig. 1. It plays an important role in tires by preventing degradation and extending its lifespan. However, when 6PPD reacts with ozone in the atmosphere, it forms a highly toxic substance called 6PPD-quinone (6PPD-Q).&nbsp;As an automotive tire wears, microscopic particles are released from the tire into the environment. These particles, tire road wear particles (TRWP), contain 6PPD-Q along with many other chemical substances. On rainy days, these particles are washed into rivers and other bodies of water, where they accumulate in the environment as microplastics.&nbsp;There are serious concerns about the negative effects these microplastics have on ecosystems and the environment. 6PPDQ has been shown to induce acute mortality in Salmonidae fish, specifically coho salmon. 1)&nbsp;Currently, there is no commercial alternative that can maintain tire stability and performance with minimal impact on the environment. Research into alternatives requires accurately assessing 6PPD levels in rubber products and objectively determining whether alternative compounds offer equivalent stability and performance while having a reduced impact on the environment.&nbsp;This article describes using a pyrolysis gas chromatograph mass spectrometer (Py-GC-MS) system to measure 6PPD levels in tire rubber.<h4>Analysis Process&nbsp;</h4>ISO/TS 21396describes a method for determining the mass concentration of TRWP in soil and sediment that uses Py-GC-MS. Similar to this method, Py-GC-MS was used for the analysis; however, this article employs optimized analytical conditions. A schematic diagram of the analysis processisshown in Fig. 2. For more information on the analysis conditions and results from the evolved gas analysis (EGA-MS, <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=42&amp;search=EGA%2FPY-3030D">EGA/PY-3030D Multi-Shot Pyrolyzer AS-1020E Auto-Shot Sampler</a>, Frontier Laboratories Ltd.) method described in Fig. 2, please see Application News No. 01-00901. The analysis conditions for the thermal desorption (TD)-GC/MS (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-QP2020+NX">GCMS-QP2020 NX</a>) method used in this article are shown in Table 1. The thermal desorption temperature was chosen based on results obtained by the EGA-MS method.<h4>Qualitative Analysis of Additive&nbsp;</h4>The total ion current (TIC) chromatogram obtained from analyzing the tire rubber is shown in Fig. 4. A spectral library search was also performed using NIST-23 and Shimadzu’s Polymer Additives Library. The library search results for the peak that eluted at around 10.43 minutes are shown in Fig. 5. Based on the results from Shimadzu’s Polymer Additives Library, the peak was estimated to be the anti-degradant Nocrac 6C (a trade name for the chemical 6PPD).<h4>Conclusion&nbsp;</h4>This article presents an analysis of the additive 6PPD in tire rubber utilizing Py-GC-MS. Good results were obtained for calibration curve linearity, calibration standard repeatability, limit of quantification, and limit of detection. Shimadzu’s Polymer Additives Library was used to identify the additive.<h3>3. Agilent Technologies: Ultrasensitive and Powerful GC/MS/MS for Analyzing Multipesticides in Black Pepper and Dried Tea</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_gcms_ms_black_pepper_5994_8783en_agilent_4b1cc85920.pdf">Full PDF for download</a></li></ul>Pesticides play a vital role in safeguarding crops and are indispensable in most farming settings to secure high yields.1 Regulatory authorities, including the Codex Alimentarius and the European Commission mandate the monitoring and reporting of pesticide residues found in or on food product.2 The analysis of pesticides in food faces several major challenges, such as the variety of pesticide classes, the complexity of different food matrices, the effects of these matrices, and the detection of target analytes at low&nbsp;concentrations.3&nbsp;According to the EU's SANTE/11312/2021 guidelines, black pepper and dried tea are considered among the difficult or unique commodity matrices for multiresidue pesticide analysis.4 The matrix interferences are complex and diverse, including carbohydrates, fats, piperine, terpenoids (for black pepper)5 , and caffeine (in dried tea). "For dried tea, matrix interferences include polyphenols, pigments, polysaccharides, alkaloids, free amino acids, high caffeine levels, and&nbsp;catechins".6&nbsp;In addition to affecting the analyte signals, matrix interferences can cause the system to become contaminated more quickly. This leads to a rapid decline in the sensitivity of the analytical instrument, and necessitates frequent maintenance, significantly reducing laboratory productivity. These are the current challenges faced by laboratories when analyzing pesticides in difficult and complex matrices.&nbsp;To overcome these challenges, Agilent offers a robust solution with its backflush and JetClean self-cleaning ion source technologies integrated into the high-sensitivity 7010B GC/TQ system. Backflush technology prevents high‑boiling contaminants from reaching the detector, reducing the need for frequent maintenance and extending column life.7 This ensures consistent performance and minimizes downtime, thereby enhancing laboratory productivity.&nbsp;With JetClean technology, your lab can significantly increase instrument uptime, as fewer manual cleanings are needed, maximizing productivity. Agilent's solution for complex matrices demonstrates the sensitivity of the 8890 GC/7010B GC/TQ and confirms its robustness when using backflush and JetClean techniques.&nbsp;<h4>Experimental and chemicals</h4><h5>GC/TQ parameters&nbsp;</h5>The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890">8890 GC</a>/<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7010B">7010B GC/TQ systems</a> (Figure 1) were optimized for wide calibration range performance. Equipped with an Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7693A">7693A automatic liquid sampler </a>and a 150-position tray, the GC operated in temperature‑programmed splitless injection mode with a multimode inlet. Detailed instrument parameters can be found in Table 1. Data acquisition was in dynamic MRM (dMRM) mode, enabling analysis of 284 pesticides with 1233 MRM transitions and up to 74 concurrent MRMs (see Figure 2). Retention time was synchronized with the Agilent Pesticides and Environmental Pollutants (P&amp;EP) 4.0 MRM Database, streamlining method&nbsp;setup.<h4>Conclusion&nbsp;</h4>This application note describes an approach for the analysis of multipesticides in complex and distinctive matrices, including black pepper and dried tea using the Agilent 8890 GC/7010B GC/TQ. The comprehensive solution successfully addresses the challenges encountered when analyzing multipesticides in difficult matrices such as black pepper and dried tea. The high sensitivity of the 7010B GC/TQ enables the application of deep dilution methods. Additionally, the use of backflush technology combined with the Agilent JetClean self-cleaning ion source ensures system stability and robustness, enhancing the operational efficiency of testing laboratories and reducing equipment downtime during&nbsp;maintenance.<h3>2. EST Analytical: Static Headspace Sampling of Volatile Organic Compounds</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/Flex_8260_by_Static_Headspace_7ddf2e4157.pdf">Full PDF for download</a></li></ul><h4>Discussion</h4>Purge and trap sampling involves purging the VOCs out of the matrix and trapping the analytes onto an analytical trap, the trap is then desorbed to the GC/MS. This process has a number of pros and cons. On the positive side, purge and trap is more sensitive. It is also the recommended sampling technique for USEPA Methods. Furthermore, the advent of autosampling systems has simplified sample preparation. However, purge and trap does have some negatives, including active sites, worries about foaming samples, and trap degradation.Static headspace sampling, on the other hand, is much simpler than purge and trap sampling. For this sampling technique, the sample is brought to equilibrium and a portion of the headspace is transferred to the GC/MS for separation and analysis. The simplicity of this technique is a definite pro. Moreover, this sampling process does not develop active sites, has no need for an analytical trap and the linear calibration range can be much higher than that of purge and trap sampling. Conversely, samples need to be manually prepped thus, losing their sample integrity. Additionally, the detection limits are higher for a number of compounds. Finally, some of the analytes do not partition into the headspace well enough and need method optimization.&nbsp;In recent years, GC/MS systems have become much more sensitive. The advent of SIM/Scan acquisition techniques has made low level detection a much simpler proposition. This analysis will focus on the headspace sampling and analysis of over 50 volatile organic compounds.&nbsp;<h4>Experimental</h4>The sampling system used for this analysis was the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/2162">EST Analytical FLEX autosampler</a> fitted with a 2.5ml headspace syringe. An <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7890+GC">Agilent 7890 GC</a> and <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=5975">5975 MS</a> were used for separation and analysis. The GC was configured with a Restek Rxi 624 Sil MS 30m x 250mm x 1.4µm column and a SKY 2mm x 6.5 x 78.5 splitless inlet liner. The MS was run in SIM/Scan mode. Refer to Tables 1 and 2 for the analysis and sampling parameters.<h4>Conclusions</h4>Static headspace sampling in conjunction with SIM/Scan acquisition proved to be a good alternative to purge and trap sampling for a number of USEPA Method 8260 compounds. The curve %RSD results showed the linearity of the curve to meet the USEPA Method 8260 requirement of 15% or better. The method detection limits of all the compounds tested also passed method requirements. Lastly, the precision and accuracy of the autosampling and analysis system was excellent, with the average precision at less than 6% RSD and the average %recovery at just over 101%.<h3>3. Shimadzu: Aromatic Component Analysis of Gasoline According to ASTM D5580 Using the Brevis GC- 2050 Gas Chromatograph</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_01024_en_039b304288.pdf">Full PDF for download</a></li></ul>Aromatics, such as toluene and xylene, are sometimes added to gasoline to improve the octane number. Measuring the levels of these aromatics is essential for quality control in gasoline production. It is also important for environmental protection, in particular, for reducing emissions of benzene, a hazardous substance, as well as for sustainable energy use that reduces the environmental impact of gasoline while maintaining its performance.ASTM D55801) is a standard test method that uses gas chromatography to measure aromatics in gasoline. Gasoline aromatics can be separated more effectively by switching between two gas chromatography columns of different polarities and venting non-aromatic components from the system. This Application News describes using the Brevis GC2050 gas chromatograph to determine aromatics in gasoline by applying ASTM D5580.<h4>Equipment Configuration and Analytical Conditions&nbsp;</h4>The system comprises a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Brevis+GC-2050">Brevis GC-2050 gas chromatograph</a>, an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=AOC-30">AOC-30</a> for sample injection, and a VB-30-S heated valve box. Aromatics were determined using two sets of Analysis Conditions that were tailored to measure different components (Table 1). Note that the timing of the flow path switching and other conditions needs to be adjusted according to the columns being used and the operating environment.<h4>Determining Aromatics in Gasoline&nbsp;</h4>A gasoline sample was prepared by adding 2 to 10 wt% aromatics and 10 wt% of an internal standard (2-hexanone) and then analyzed under Analysis Conditions A and Analysis Conditions B. Fig. 2 shows the chromatograms obtained under each set of Analysis Conditions. Despite gasoline containing large amounts of hydrocarbons, oxygenates, and other impurities, these impurities did not appear as peaks in the chromatograms, and only the aromatics added to the gasoline were detected. These results verify the utility of using flow path switching to effectively utilize the characteristics of two different columns.<h4>Conclusion&nbsp;</h4>A flow path switching valve was installed on a Brevis GC-2050 gas chromatograph to enable the determination of gasoline aromatics (benzene, toluene, ethylbenzene, p/m-xylene, oxylene, and C9+ aromatics) according to ASTM D5580.&nbsp;This system can switch between different columns during analysis to determine specific aromatics from among the many compounds present in gasoline. Analysis was performed under two sets of conditions, each using different flow path switching times. This method met the requirements for linearity and reproducibility described in ASTM D5580. The timing of flow path switching can also be adjusted to allow the targeted determination of oxygenates in gasoline. The ability to perform multiple analyses on a single system means the Brevis GC-2050 offers even greater functionality as a compact gas chromatograph.

Agilent Technologies: Volatile PFAS in Cosmetics Using PAL3 Coupled with Triple Quadrupole GC/MS</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_pfas_cosmetics_pal3_gc_tq_5994_8752en_agilent_52b56afd36.pdf">Full PDF for download</a></li></ul>Per- and polyfluoroalkyl substances (PFAS) are a large family of synthetic organic chemicals containing carbon chains where most or all the hydrogen atoms are replaced by fluorine. Due to the strength and stability of the carbon‑fluorine bond, PFAS tend to be very stable and chemically inert, which makes them both very useful in various applications and resistant to degradation. This persistence causes environmental contamination and bioaccumulation in humans and wildlife; as a result, PFAS are often labeled "forever chemicals".1&nbsp;In cosmetics, PFAS are valued for their functional properties, including emulsification, stabilization, and water or sweat resistance. Despite these benefits, concerns have grown over their safety due to direct exposure from repeated use through skin, eyes, and mouth. The OECD's 2024 report underscores the risks associated with PFAS in cosmetic formulations, particularly given their potential for long-term health and environmental impacts.2,3 Regulators around the world are taking decisive steps to ban or restrict PFAS in cosmetics. For example, in the EU, various types of PFAS have been regulated under REACH and POPs regulations that span most industries, including cosmetics.4,5 EU Cosmetics Regulation (EC) No. 1223/2009 has listed the five types of PFAS as the substances prohibited.6 A broader EU-wide ban is underway, with France having passed its own ban on PFAS in cosmetics effective 1 January 2026, as part of a larger proposal to ban PFAS in consumer products.7,8 The US has not banned the use of PFAS in cosmetics at the federal level yet. The USFDA has been evaluating the use of PFAS in cosmetics under the authority of the Cosmetic Regulatory Modernization Act (MoCRA), and is expected to release a report on safety and risk by the end of 2025.9 Although the federal government is still on the sidelines, many states in the US have been rapidly enacting or implementing the ban and reporting requirements for PFAS in cosmetics, with effective dates ranging from 2025 to 2032.10-13 Canada has proposed a ban on the use of PFAS in various consumer products, including cosmetics.14 South Korea has added 12 PFAS into a restricted substances list for cosmetic products in 2024.15 The Environmental Protection Authority (EPA) of New Zealand has officially announced a ban on the use of PFAS in cosmetic products, effective from 31 December 2026.16&nbsp;As regulations surrounding PFAS continue to evolve, developing a sensitive and robust analytical method is crucial to meet stringent guidelines. The objective of this study was to demonstrate a fully automated workflow using PAL3&nbsp;Series&nbsp;2 RTC autosampler coupled with an Agilent 7010D triple quadrupole GC/MS to quantify more than 30&nbsp;volatile PFAS analytes in cosmetics. The automation method performance includes sensitivity, accuracy, and precision studies.<h4>Experimental</h4><h5>Instrumentation&nbsp;</h5>An <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890+GC">Agilent 8890 GC </a>with a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7010D">7010D GC/TQ</a> equipped with HES 2.0 ion source, coupled with a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=PAL3">PAL3</a> Series 2 RTC autosampler, was used for this analysis. An MMI inlet and splitless liner (part number 5190-2293) were used and chromatographic separation was performed using an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=J%26W+DB-624">Agilent J&amp;W DB-624</a>, 30 m × 0.25 mm, 1.40 μm column (part number 122-1334UI). The instrument setup is shown in Figure 1. The acquisition method for 34 native PFAS and four ISTDs, including GC condition, TQ parameters were described in a previous application note.17<h4>Sample analysis&nbsp;</h4>Seven cosmetic samples from various brands—including foundations, lipsticks, and mascaras—were analyzed. One liquid foundation was designated as a quality control (QC) sample, while the remaining six were treated as unknowns. According to the OECD report published in 2024, cosmetic products such as shampoo, eye makeup, foundation, facial cleansers, lipsticks, and lip glosses may contain PFAS, which serve functional or performance-enhancing roles.2,19 In this study, several PFAS compounds were detected above the method detection limit (MDL) in the unknown samples, including 10:2 FTOH, MeFHxSA, MeFOSE, EtOSE and 1H,1H‑perfluorooctyl acrylate. The results also demonstrated that the automated workflow is effective for PFAS extraction and quantitation in cosmetic matrices.<h4>Conclusion&nbsp;</h4>An accurate and robust automated workflow was successfully developed using the PAL3 Series 2 RTC autosampler coupled with the Agilent 7010D GC/TQ system for the analysis of volatile PFAS in cosmetic products. Most PFAS targets achieved an LOQ of 50 µg/kg and MDL below 1.5 µg/kg, with recoveries falling within the acceptable range of 60% to 110%. These results demonstrate excellent extraction efficiency and reliability of the automated workflow. Furthermore, automation significantly reduces human error and enhances productivity, while maintaining high sensitivity in PFAS analysis.<h3>2. Shimadzu: Simplifying Multicomponent Quantitative Analysis of Organic Compounds with the Polyarc Microreactor for GC</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_00945_en_9f7adeedc4.pdf">Full PDF for download</a></li></ul><h4>User Benefits&nbsp;</h4><ul><li>The Polyarc system converts organic compounds eluting from the column into methane, enabling sensitivity to be normalized to carbon concentrations.&nbsp;</li><li>Even for compounds containing heteroatoms or unsaturated bonds, which tend to show lower sensitivity with FID, the Polyarc reduces response differences.&nbsp;</li><li>Because the response is normalized, fewer calibration curves are required even for multicomponent sample analysis.</li></ul>The Flame Ionization Detector (FID), the most widely used detector for GC, is used for quantitative analysis across a wide range of fields because it can detect almost all organic compounds. However, FID can exhibit sensitivity differences depending on the compound type, such as compounds containing heteroatoms (e.g., oxygen or nitrogen) or those with unsaturated bonds (for details, refer to Application News No.01-01033-EN). As a result, conventional quantitative methods using calibration curves— including external standard methods— typically require a separate calibration curve for each target analyte. This application introduces a case study analyzing a mixed solution of eleven compounds with different functional groups using the Polyarc, a GC microreactor. While compounds exhibit varying sensitivity with FID detection, the Polyarc system reduces sensitivity differences between compounds and facilitates quantitative analysis through simplified calibration curve creation.<h4>Overview of the Polyarc</h4>The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?search=Polyarc">Polyarc</a> is a post-column microreactor installed between the column and the FID. The main unit is mounted on the GC’s top panel. A schematic isshown in Fig. 1. Organic compounds introduced into the Polyarc via the column are fully converted to methane in two steps—oxidation followed by reduction (Fig. 2)—and are ultimately detected by FID. Because the FID detects methane, sensitivity is determined by the number of carbons, regardless of the original organic compound’s functional groups. For example, 1 mol of 1- propanol (C3H8O) is detected as 3 mol of methane.<h4>Analytical Conditions&nbsp;</h4>In this application, the sample was prepared by dissolving nheptane, cyclohexane, toluene, o-xylene, methyl ethyl ketone (MEK), tetrahydrofuran (THF), N,N-dimethylformamide (DMF), butyl acetate, propylene glycol monomethyl ether acetate (PMA), pyridine, and 1,2-dichloroethane in methanol so that each compound was 500 ppm (v/v%). Analytical conditions are shown in Table 1. The instrument used was a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Nexis+GC-2030">Nexis GC-2030</a> equipped with the Polyarc Ultra was used.(Fig. 3) Polyarc can be installed on both the Nexis GC-2030 and the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Brevis+GC-2050">Brevis GC-2050</a>. Additionally, in this application, N2 was used as the carrier gas for affordability and safety, enabling cost-effective analysis.<h4>Conclusion&nbsp;</h4>Analyzing multicomponent samples with a Polyarc confirmed that sensitivity differences due to functional groups can be reduced. Compounds containing heteroatoms or unsaturated bonds can be analyzed with relative response comparable to hydrocarbons when a Polyarc is used. Peak area reproducibility was excellent, with %RSD &lt; 1. Additionally, it was confirmed that analyzing at least one compound of known concentration enables quantitation of compounds with unknown concentration—i.e., calibration curve preparation can be omitted.Table 5 summarizes the benefits of using a Polyarc system. The Polyarc simplifies multicomponent quantitative analysis and enables high-sensitivity analysis of compounds that are poorly detected by FID. Note that for certain compounds, adsorption in the inlet or similar effects may prevent achieving a relative response equivalent to that of saturated hydrocarbons.<h3>3. They provide a safe, hygienic and non-judgement environment in which individuals may consume illicit drugs without fear of arrest.&nbsp;The benefits of SIS are to:&nbsp;(1) prevent an overdose from becoming fatal through quick intervention, (2) reduce the frequency of public injection and publicly discarded syringes, (3) increase access to health services including substance use treatment, and (4) change behaviors associated with acquiring HIV and hepatitis.&nbsp;The benefits of drug checking services within an SIS are:&nbsp;(1) it provides the consumer information about the contents of their drugs prior to consumption allowing them take steps to reduce their risk of overdose, (2) it identifies when a highly toxic substance may be in the local drug supply so advisories can be issued to local service agencies and first responders, (3) it encourages use of supervised injection services and the benefits mentioned above, and (4) it detects novel substances or drug combinations that may impact the consumer.&nbsp;Initially approved by the Canadian federal government in 2017, the nation now has four official SISs in operation in Ottawa and four sites approved in Toronto. It is projected to reach approximately $173 billion by 2030, implying a steady growth rate of roughly 4% [1]. This growth is driven by sustained consumer demand and is expected to continue rising.&nbsp;Spectroscopy is increasingly utilized in chocolate manufacturing for quality control (QC), thanks to its ability to assess the composition of chocolate, providing a «fingerprint» spectrum that reveals its chemical details. Specifically, Raman can be used in QC to distinguish between types of chocolate, detect adulteration, measure crystallization and texture, and monitor the manufacturing process.&nbsp;This Application Note outlines techniques for effectively collecting Raman spectra from various chocolates, providing a foundation for quality assessment and adulteration detection.<h4>METHOD&nbsp;</h4>Raman data collection with an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=220&amp;search=i-Raman+NxG+1064">i-Raman NxG 1064 </a>laboratory Raman system (Figure 2) was optimized by adjusting integration time and laser power (Table 3) to determine the best conditions to maximize signal strength and minimize risk of sample melting.All chocolate samples were analyzed by placing a piece of chocolate on the stage with a Raman probe securely locked above the sample (Figure 3). The optimal working distance was determined by adjusting the probe’s z-axis position while continuously monitoring the intensity of the Raman signal. Once the optimal focal distance is found, a distance regulator helps the operator position the probe on the sample to ensure consistent and reliable measurement.<h4>CONCLUSION</h4>This study highlights the capability of Raman spectroscopy for rapid, nondestructive measurements of chocolate quality indicators. PLS models demonstrate high predictive accuracy for both cocoarelated materials and sugar content. Increasing the number of samples and testing a broader range of chocolates would further improve the robustness and accuracy of the model. Overall, Raman spectroscopy, combined with chemometric modeling, offers a reliable QC method for routine and real-time chocolate analysis.<h3>4. The act does not regulate plastic but rather promotes the establishment of a sustainable circular economy by encouraging collaboration among businesses, local governments, and consumers throughout the overall process of designing, manufacturing, selling, collecting, and recycling plastic products.&nbsp;One of these initiatives is the recycling of PET bottles for beverage use. When recycling beverage bottles, residual odors can be an issue, with substances like acetaldehyde from water bottles and limonene from citrus beverages known to persist in the containers. To address these residual substances, recycling companies are implementing unique strategies, one of which involves using gas chromatography-mass spectrometry (GC-MS) to quantify the residual materials. In this article, we present an example of qualitative and quantitative analysis of aldehydes and limonene in PET bottles using the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-QP2020+NX">GCMS-QP2020 NX</a>/HS-20 NX, with hydrogen (H2) as the carrier gas.<h4>Conclusion</h4>By using hydrogen (H2) gas as the carrier gas, we were able to avoid the use of the hard-to-obtain helium gas. Additionally, by employing an HS-GC-MS system, we could qualitatively and quantitatively analyze acetaldehyde and limonene from PET without the need for cumbersome pretreatment.

Microplastics are generally defined as plastic particles with a maximum dimension of ≤ 5 mm, while nanoplastics typically range from 1 to 1000 nm in size. <a href="javascript:void(0);">(2)</a> NMPs encompass a wide variety of polymer particles that originate either from direct manufacture (e.g., microbeads and synthetic textiles) or from environmental degradation processes (e.g., photooxidation and mechanical abrasion) of larger plastic debris (i.e., macroplastics, &gt;5 mm). <a href="javascript:void(0);">(3)</a>NMPs are ubiquitous in the environment. They have been detected from Antarctic coast sediments to Arctic ice sheets, the summit of Mt. Everest to the depths of the Mariana Trench, and have been found in organisms across all trophic levels, including humans. <a href="javascript:void(0);">(4−10)</a> The pervasive presence of NMPs has raised significant ecological and human health concerns. NMPs can act as vectors for environmental pollutants. Hydrophobic organic contaminants (e.g., PCBs and PAHs) readily adsorb onto their surfaces, and plastics additives such as phthalates and flame retardants can leach into surrounding media (e.g., water, sediment, tissue). <a href="javascript:void(0);">(11)</a> Even without associated harmful chemicals, the physical presence of NMPs can lead to increased production of reactive oxygen species in cells, resulting in an inflammatory response. <a href="javascript:void(0);">(12)</a> Given the widespread NMP pollution and its complex ecological ramifications, there is a critical need for more robust high-throughput quantitative methods to accurately assess NMPs in diverse matrices.Numerous analytical techniques have been employed for the detection and quantification of NMPs, which often combine traditional microscopy with advanced spectroscopic and molecular approaches. <a href="javascript:void(0);">(13)</a> These methods yield either estimates of particle counts or mass-based concentrations normalized to sample volume or weight. Prior to analysis, most methods require extensive chemical or enzymatic digestion and subsequent cleanup steps to isolate plastic particles and reduce matrix complexity. <a href="javascript:void(0);">(14,15)</a> Particle count-based methods encompass optical microscopy, laser diffraction particle size analysis, scanning electron microscopy, flow cytometry, and Fourier-transform infrared (FTIR) and Raman spectroscopy. <a href="javascript:void(0);">(16)</a> These methods provide quantitative estimates of particle abundance across the entire size spectrum of NMPs, and some can also differentiate between plastic types.Thermal decomposition coupled with gas chromatography–mass spectrometry (GC-MS) is an increasingly utilized technique for the analysis of NMPs. <a href="javascript:void(0);">(17,18)</a> Pyrolysis-based methods thermally decompose plastic polymers in samples under oxygen-free conditions at temperatures exceeding 500 °C generating characteristic volatile fragments. These fragments are then separated by gas chromatography and detected by mass spectrometry with high sensitivity and specificity. As with other analytical techniques, pyrolysis-GC-MS (PyGC-MS) performance can be improved by incorporating sample cleanup steps prior to analysis to remove interfering organic and inorganic constituents. Matrix interference can often lead to misidentification of plastic polymer types and disrupt the pyrolysis process, ultimately compromising analytical accuracy and precision. <a href="javascript:void(0);">(19)</a> For example, lipids in biological tissues can yield similar combustion products to those of polyethylene (PE), polypropylene (PP), and nylon-66 (N-66). <a href="javascript:void(0);">(20)</a>Improvements that enable more reliable quantification of NMPs at ultralow concentrations by PyGC-MS include optimized sample treatment methodologies that include the addition of reactants to improve pyrolysis efficiency of plastic polymers, and the application of advanced mass spectrometric methods. Quantification of NMPs at trace levels is often constrained by the minimum weight limits of analytical balances, as calibration relies on the mass of particulate standards. This limitation has been addressed by using inert fillers, such as calcium carbonate (CaCO3) or clean quartz sand that are mixed with plastic standards to produce weighable calibration standards down to the nanogram level. While single-quadrupole mass detectors are widely used due to their simplicity and low cost, they offer limited sensitivity and are less sensitive in complex matrices. <a href="javascript:void(0);">(21)</a>The application of time-of-flight and orbitrap mass spectrometry, as well as modern triple-quadrupole mass detectors, promises improved sensitivity and specificity of NMP quantification in complex matrices. These highly selective detectors enable precise mass determination, reducing ambiguity in peak identification and enhancing the ability to differentiate plastics from natural organic matter commonly present in environmental samples. Time of flight and orbitrap mass spectrometers support untargeted screening and identification of emerging or unknown plastic-related compounds, while triple-quadrupole instruments excel in targeted quantification with high throughput and reproducibility, which are advantageous features for routine monitoring. <a href="javascript:void(0);">(22−24)</a> Compared to traditional methods such as FTIR or Raman spectroscopy, modern mass spectrometric approaches are better equipped to address the challenges posed by complex sample matrices, diverse plastic compositions, and trace-level concentrations, making them essential tools for future NMP analysis. <a href="javascript:void(0);">(25,26)</a>Here we present a novel analytical method for the identification and quantification of NMPs in diverse environmental matrices that include marine waters, sediments, and biological tissues by using pyrolysis gas chromatography coupled with triple quadrupole mass spectrometry (PyGC-qQq-MS). Depending on the sample matrix, NMPs were concentrated through filtration, extraction, or digestion prior to analysis. Gas chromatography with qQq-MS detection using dynamic multiple reaction monitoring (DMRM) mode with an internal standard yielded significantly improved selectivity and sensitivity, enabling accurate and precise quantification of plastic polymers. Method validation demonstrated the robust performance of the method across diverse sample types, with reliable detection of NMPs at amounts of 1–126 ng and in the presence of significant matrix interference.<h4>Experimental Section</h4><h5>Pyrolysis GC/MS qQq-MS Analysis</h5>Pyrolysis gas chromatography was carried out with a Frontier Auto-Shot AS-2020E sampler and an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=42&amp;search=EGA%2FPY-3030D">EGA/PY-3030D multishot pyrolyzer</a> connected to an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890+gc">Agilent 8890 GC</a> and <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=7010B">7010B qQq-MS</a>. Prior to pyrolysis at 600 °C, GF/F with particulates from water samples or digested tissues were spiked with PFS, which served as an internal standard. Filters were then transferred to Frontier Eco Cups and topped with 5 mg of CaCO3 and quartz wool to ensure proper packing. Redissolved sediment extracts were transferred directly into the Eco Cup LFs, dried under a gentle stream of N2, and packed with CaCO3 and quartz wool.<h4>Results and Discussion</h4><h5>Pyrograms</h5>The method was optimized to yield characteristic pyrolytic products of plastic polymers that were well resolved on a Frontier Ultra Alloy 5 column with a linear temperature program. Pyrograms of surface water, vertebrate and invertebrate tissue, and sediment samples, alongside a standard plastic mixture are shown in <a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/10.1021/acs.analchem.5c05604#fig3">Figure 3</a>. Pyrolysis temperature was optimized to ensure efficient volatilization of polymers, and specific product ions were chosen to minimize matrix influence that can generate pyrolysis products with similar mass spectra. For each polymer, precursor and product ions were selected to minimize interference from natural polymers and characteristic fragmentation reactions. For polymers that primarily undergo depolymerization (e.g., PE, PP, PS, PMMA), the selected precursor ions corresponded to characteristic oligomeric or monomeric pyrolysis products, and the quantifier ions were derived from simple bond cleavages or rearrangement reactions. For example, PS pyrolysis products included mono, di-, and trimeric units with different retention times. Characteristic and abundant molecular precursor ions were identified for each plastic polymer and evaluated against natural polymers to minimize matrix interference. Fragmentation ions derived from these precursor ions were selected based on characteristic fragmentation reactions. The more abundant product ion was used for quantification, whereas the secondary ion was employed as the qualifier to monitor potential coelution with matrix compounds. Fragment losses usually corresponded to the loss of polymer subunits or dehydration reactions. Operating in DMRM mode improved sensitivity by maximizing acquisition time for each targeted analyte. Ratios of quantifier to qualifier ion (<a target="_blank" rel="noopener noreferrer" href="https://pubs.acs.org/doi/suppl/10.1021/acs.analchem.5c05604/suppl_file/ac5c05604_si_001.pdf">Table S2</a>), together with retention times, ensured both compound identification and confirmation.<img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Anal_Chem_2026_98_1_633_641_Table_1_Plastic_Polymer_Abbreviations_d796d20f8b.jpg" alt="Anal. PFS was dissolved in toluene and added to the sample prior to pyrolysis. The use of deuterated polystyrene as an internal standard was explored; however, extensive hydrogen–deuterium exchange occurred during pyrolysis, leading to pyrolysis products with variable D/H content. <a href="javascript:void(0);">(31)</a> Similarly, attempts to use brominated polystyrene were unsuccessful, as bromine was readily lost from the polymer chain during pyrolysis, resulting in products indistinguishable from those of PS and others.Calibration at &lt;1 mg plastic mass was accomplished using plastic standards dispersed in CaCO3. The most consistent results were achieved with commercially available standard kits (e.g., Frontiers), which contain homogeneously distributed plastic polymers in CaCO3. Attempts to prepare homogeneous mixtures of plastic polymers in CaCO3 by ball milling finely ground plastics with CaCO3 yielded less consistent results than commercial standards.<h4>Conclusion</h4>This study developed and validated a robust analytical workflow for the detection and quantification of NMPs across a diverse range of environmental matrices. The method demonstrated high sample throughput, robust compensation of matrix interference, and high precision and accuracy. This was accomplished through efficient sample cleanup procedures, internal standard calibration, tandem mass spectrometry, and a correction for lipid interference in the detection of PE, PP, and N-66. The applicability of the method was demonstrated on marine surface waters, sediments and soils, and muscle tissue of fishes and oysters. Results revealed quantifiable concentrations of plastics in all sample types emphasizing the pervasive and persistent nature of plastics in the environment and their bioaccumulation potential across trophic levels. This method provides a critical tool for studying NMPs in the environment, ecological risks, and potential impacts on human health.

Agilent Technologies: Total Hydrocarbon Impurity Analysis in PEM Fuel Cell Grade Hydrogen Using the Agilent 8890 GC-FID System</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_hydrocarbon_impurity_analysis_pem_fuel_cell_8890_gcfid_5994_8935en_agilent_00a26949ef.pdf">Full PDF for download</a></li></ul>High-purity hydrogen is crucial for the optimal and long-term performance of fuel cells.&nbsp;Different than the industry grade hydrogen used for chemicals, refining, and metal processing, where small amounts of impurities are acceptable. Fuel cell hydrogen is required to meet the species-specific impurity limits described in the specifications. Such impurities include hydrocarbons, sulfur-containing compounds, ammonia, permanent gases, halogens, and formaldehyde. Each group of compounds has its own regulation limit. The limits are defined based on their impact on the fuel cell lifetime and performance. ISO 14687 and SAE J2719 are the most recognized standards for regulation of fuel cell hydrogen quality.&nbsp;Multiple testing points are implemented throughout the whole supply chain of fuel cell grade hydrogen—from its production, storage, and distribution to its final application. The analytical approaches employed depend on the impurity type and the corresponding limit. Among the varied contaminants, hydrocarbon impurity is controlled based on THC amount (excluding methane because it has a separate specific limit) and the quality limit as specified in ISO 14687 and SAE J2719 (2&nbsp;ppm).&nbsp;There are two ways to analyze THCs in hydrogen. One is to separate and detect each hydrocarbon, then add their concentrations to get the THC amount (excluding methane). This analytical approach is recommended by the GB/T 44238-2024 test method.1 The other way is by eluting all hydrocarbons as a single peak and quantitating the peak according to methane's response factor (RF). The total THC is then obtained by subtracting the methane amount, which is analyzed by a separate method.In this application note, THC impurity was tested using the second approach. An Agilent 8890 GC with FID system were used for THC analysis, and the Agilent <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=990+Micro+GC">990 Micro GC</a> was used for methane measurement. The methane analysis solution is published in application note 5994-8830EN.2 Here, we will showcase THC analysis using the 8890 GC-FID system.&nbsp;<h4>Experimental&nbsp;</h4>The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890">8890 GC</a> is configured with one gas sampling valve, a split/splitless (S/SL) inlet, and a flame ionization detector (FID), as shown in Figure 1. The fixed volume of gas sample is injected into the S/SL inlet and part of the sample flows into an uncoated capillary column, then elutes off as a single peak for detection. Table 1 lists experimental conditions for the analysis. The standard gases shown in Table 2 are used to verify system precision, generate methane's RF, and calculate method detection limit (MDL).<h4>Conclusion&nbsp;</h4>This application note demonstrates THC&nbsp;analysis in fuel cell hydrogen using the Agilent 8890 GC-FID system. The hydrocarbons eluted as a single peak on an uncoated column and were quantitated using methane's RF. The method repeatability and reproducibility were around 2%. Method accuracy was better than 120%. The MDL was 0.97&nbsp;ppm. All results met the analytical requirements of regulating THC in fuel cell grade hydrogen according to SAE&nbsp;J2719 and ISO 14687.&nbsp;The applied analytical approach requires methane analysis by a separate method, which can be realized using the Agilent 990 Micro GC or using a coated column followed by FID detection on another channel of the 8890 GC system.<h3>2. BaySpec: Rapid Detection of Trace Levels of Deoxynivalenol in Wheat Flour by Portable Miniature Linear Ion Trap Mass Spectrometry</h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/Bay_Spec_App_Note_MS_Portability_In_Wheat_Quality_04959a96bd.pdf">Full PDF for download</a></li></ul>Deoxynivalenol (Cas:51481‐10‐8), also known as DON, is a trichothecene mycotoxin that is widely found in grains such as wheat, barley, and rice.&nbsp;&nbsp;The molecular structure of DON is provided in figure 1.&nbsp;&nbsp;DON is linked to two plant pathogens: Fusarium graminearum (wheat and corn), and Fusarium ear blight (corn only).&nbsp; &nbsp; DON inhibits the synthesis of DNA / RNA as well as protein synthesis at the ribosomal level, ultimately resulting in the loss of yield and contamination of seeds.&nbsp;&nbsp;An acute dose of DON can induce vomiting (emesis) in pigs, whereas at lower concentrations in the diet, it reduces growth and feed consumption (anorexia).1&nbsp;The U.S. FDA has established DON advisory levels to provide safe food and feed at concentrations of &lt; 10 ppm to &lt; 1 ppm depending on application.2Because DON is difficult to derivatize, this compound is typically analyzed via thin‐layer chromatography (TLC) or high performance liquid chromatography (HPLC).3 While TLC is simple, fast, and economical, this analytical technique does not provide high accuracy nor consistent results on a day‐to‐day basis.&nbsp;&nbsp;HPLC improves on accuracy and consistency but requires large quantities of costly solvents and produces large volumes of hazardous waste.&nbsp;&nbsp; Depending on sampling requirements and system performance, the annual maintenance costs for a standard HPLC instrument can run ~$10‐20k per year.4 &nbsp;&nbsp; Furthermore, although HPLC systems can be easy to use when properly configured, a trained professional is typically needed for troubleshooting, maintenance, and developing new methods.Two samples were analyzed in a recent case study for an agricultural firm based in Canada.&nbsp;&nbsp;&nbsp;One sample contained 200 mg of wheat powder, while the other sample was 1 mg of the pure DON compound. Measurements were taken on a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/2219">BaySpec PortabilityTM Mass Spectrometer</a> (Figure 2) with both thermal desorption (TD) electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI) methods. Samples were tested “dry” either by probing the surface of the sample vials or by spreading the powder onto a clean surface and swabbing. An enlarged image of the sampling probe with a small amount of wheat powder is shown in Figure 3.<h3>3. MDCW / JEOL: Structural elucidation using GCxGC-TOFMS and machine learning for unknown metabolites in HeLa cell</h3><ul><li>Presentation</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/O_7_Masaaki_Ubukata_2_56a67fdf00.pdf">Full PDF for download</a></li></ul>The presentation by Masaaki Ubukata focuses on advancing structural elucidation of unknown metabolites in HeLa cells using comprehensive two-dimensional gas chromatography coupled with high-resolution time-of-flight mass spectrometry (<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?instrumentationItems=2-13-3-5&amp;mainauthorItems=23">GC×GC-HRTOFMS</a>) and machine learning. The work addresses a major limitation in metabolomics: while GC-MS offers highly reproducible EI fragmentation and strong library support, only a tiny fraction of known chemical space is represented in standard databases such as NIST (about 0.3% of compounds). To overcome this gap, the team developed an AI-based approach capable of predicting EI mass spectra and retention indices for over 200 million compounds, creating a large in-silico spectral library for unknown identification.The workflow integrates accurate mass measurements from GC-HRTOFMS with both electron ionization (EI) and soft ionization data. Molecular formulas are first determined using isotopic patterns and fragment ion accuracy, then candidate structures are narrowed using predicted retention indices and compared against AI-predicted EI spectra ranked by cosine similarity. Model performance has steadily improved, reaching an average cosine similarity of 0.86, with correct structures ranked first in 56% of evaluation cases and within the top 10 in 87% of cases. This demonstrates strong capability for resolving unknown compounds beyond conventional library searches.The approach was applied to HeLa cell metabolomics, a well-established human cell model. Using GC×GC separation, more than 800 metabolites were detected. Importantly, the system successfully identified N-methyl-uridine monophosphate (N-methyl UMP), a biologically relevant RNA-modification-related metabolite not registered in the NIST database. After molecular formula determination and AI ranking, N-methyl UMP was identified as the top candidate among 53 structural isomers sharing the same formula, demonstrating the practical strength of combining high-resolution MS, soft ionization, and AI-based spectral prediction.In conclusion, the study highlights the synergistic power of three technologies: high-resolution GC-TOFMS, soft ionization techniques, and AI-driven structural analysis. Together, they significantly enhance the identification of unknown metabolites in non-targeted metabolomics and provide a scalable solution for exploring chemical space far beyond existing experimental spectral libraries.<h3>4. These aroma components vary based on the type and variety of the fruit, its ripeness, production area, and cultivation environment. The complex interplay of these factors shapes the unique flavors of individual fruits. Therefore, the analysis of aroma components plays an important role in quality control and the development of new fruit varieties. The compounds that make up the aroma of fruits are diverse, including both volatile and non-volatile components, and their numbers can range from dozens to hundreds. These components, which include alcohols, esters, ketones, and acids, possess various chemical structures and contribute to the aroma of the fruit. Gas chromatography mass spectrometry (GC/MS) offers an effective method for objectively evaluating the relationship between aroma and aroma components. This article describes an example of analyzing the aroma components in apples using a <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMS-QP2050">GCMS-QP2050</a> gas chromatograph mass spectrometer in combination with an<a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=HS20+NX"> HS20 NX headspace sampler</a> (Fig. 1).<h4>Multivariate Analysis&nbsp;</h4>Three types of apples, with five samples each (for a total of 15 samples), were measured once. The obtained data were analyzed using SIMCA17 multivariate data analysis software (Informatics Co., Ltd.) to perform principal component analysis (PCA) and hierarchical clustering analysis. The PCA score plot (Fig. 2), loading plot (Fig. 3), and the dendrogram from the hierarchical clustering analysis (Fig. 4) are shown.&nbsp;Based on the PCA score plot, the contribution rate of the first principal component (horizontal axis) is 85.6 %, and the contribution rate of the second principal component (vertical axis) is 10.2 %, totaling 95.8 %. The clear separation of the plots by apple type indicates that there are differences in the aroma of the three varieties. Additionally, the results of the hierarchical clustering analysis show that Jonagold (A) and (B) have relatively similar compositions of aroma components.&nbsp;Table 3 shows the aroma components that are relatively abundant in Jonagold (A) along with the sensory information for each aroma component registered in the Smart Aroma Database.<h4>Conclusion&nbsp;</h4>We analyzed the aroma components of three types of commercially available apples using a GCMS-QP2050 gas chromatograph mass spectrometer. By utilizing an HS-20 NX headspace sampler, we were able to analyze the aroma components of fresh apples with high sensitivity, and the Smart Aroma Database made the process convenient.&nbsp;We consider that scientifically analyzing the differences in aroma components based on factors such as fruit type, variety, ripeness, production area, and cultivation environment is beneficial for quality management and the development of new varieties.
