<div><p><strong>Per- and polyfluoroalkyl substances (PFAS) represent a vast class of more than 3,000 synthetic compounds containing perfluorinated chains (with the general formula CnF₂n+₁COOH), commonly utilized in a wide range of consumer products and industrial processes.</strong></p><p>The unique structure of PFAS, characterized by multiple carbon-fluorine (C-F) bonds, contributes to their <strong>exceptional chemical stability and resistance</strong> to degradation through environmental or biological processes. This durability makes them some of the <strong>most persistent pollutants in the environment</strong>. Their resistance to water and oil has led to widespread use in diverse applications, including water- and stain-repellent coatings for textiles, paints, personal care items, firefighting foams, fluoropolymer-based lubricants and additives, as well as grease- and water-resistant food packaging. Due to their extensive use, PFAS have been found in various environmental and biological samples, such as <strong>food, drinking water, wastewater, and human blood</strong>.</p></div><div><div><div><div><h3><strong>Why PFAS Testing Is Crucial?</strong></h3></div></div></div></div><div><p>Like many other chemicals, PFAS can cause<strong> </strong>various <strong>harmful health effects</strong>, depending on factors such as the level, duration, and route of exposure, as well as individual characteristics like age, sex, ethnicity, overall health, and genetic background. PFAS exposure has been linked to various health effects in both sexes, with some impacts supported by high certainty evidence—such as <strong>thyroid disease, liver damage, kidney cancer, and testicular cancer</strong>.</p><p>In females, PFAS may also affect pregnancy outcomes and fetal development, including increased miscarriage risk, reduced birth weight, and delayed mammary gland development. Because of their toxic nature, these compounds have been classified as “persistent organic pollutants” by the<strong> Stockholm Convention</strong> (including PFOS, PFOA, PFHxS, and their salts) and by the <strong>European Commission</strong> (including PFOA, PFOS, PFNA, and PFHxA). Their durability, chemical stability, harmful effects, and tendency to build up in living organisms make them a serious risk to both environmental and human health; hence, PFAS testing is crucial for monitoring and mitigating these threats.</p></div><div><div><div><div><h3><strong>PFAS Testing Techniques</strong></h3></div></div></div></div><div><p>PFAS testing employs advanced techniques to detect, identify, and quantify both known and emerging compounds across diverse environmental matrices.</p><h5><strong>Liquid Chromatography-Mass Spectrometry (LC-MS/MS)</strong></h5><p>LC-MS is a <strong>commonly used PFAS testing method</strong>. Samples are first processed using suitable preparation techniques, such as <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products?category=136&amp;manufacturerItems=50"><strong>solid phase extraction </strong></a>(SPE). The resulting extracted samples are then examined using LC/MS/MS with calibration performed via standards. Target analytes are identified with help of isotopically labeled surrogate standards. . Additionally, product ion ratios are compared with those from standards to support identification. Once confirmed, the concentration of each identified target compound is determined based on the response of its main product ion using external standard calibration.</p></div><div><div><div><div><h4><strong>Gas Chromatography-Mass Spectrometry (GC-MS)</strong></h4></div></div></div></div><div><p>Non targeted analysis of PFAS has largely been dominated by LC-MS methods, although GC-MS can access a complementary chemical space, especially for volatile and semivolatile PFAS. Studies show that LC‑MS covers most ionic PFAS; GC‑MS captures neutral/volatile precursors less amenable to LC, highlighting the need for GC-based approaches. GC is particularly valuable for identifying novel PFAS and transformation products from sources like manufacturing waste, incineration, wastewater, and consumer products.</p></div><div><div><div><div><h4><strong>Total Organic Fluorine (TOF)</strong></h4></div></div></div></div><div><p>Total Organic Fluorine (TOF) analysis is an indirect method used to estimate the overall burden of per- and polyfluoroalkyl substances (PFAS) and other organofluorine compounds in a sample. Instead of targeting individual PFAS molecules, TOF methods quantify the sum of organically bound fluorine.</p><p>Because this process measures the organic fluorine present within a sample, it provides a helpful bulk measurement of fluorine from all PFAS present, including those which may not be covered by targeted LC-MS/MS methods.</p></div><div><div><div><div><h4><strong>High Resolution Mass Spectrometry (HRMS)</strong></h4></div></div></div></div><div><p>HRMS is increasingly used to detect emerging PFAS, with two main strategies applied for identification: suspect screening (SS) and non-target screening (NTS). SS is a more direct method, particularly effective when tandem mass spectrometry (MS/MS) reference spectra are available. In contrast, NTS is critical for uncovering new PFAS compounds in the absence of a suspect list, using fragment-based or homologue-based techniques.</p><p>HRMS contributes significantly by offering precise mass measurements, which are essential for identifying unknown substances. Since PFAS often form homologous series, patterns in accurate mass, retention time, and common fragments (such as CF₂, C₂F₄, and CF₂O) help in their recognition.</p></div><div><div><div><div><h3><strong>Choosing the Right Testing Method</strong></h3></div></div></div></div><div><p>C-MS/MS is the primary technique used for targeted PFAS analysis due to its high sensitivity and accuracy. It is validated by the U.S. EPA through established methods such as 1633, 533, and 537.1 for detecting PFAS in various environmental and drinking water samples. For laboratories seeking reliable and compliant workflows, choosing a comprehensive <a target="_blank" rel="noopener noreferrer" href="https://www.phenomenex.com/industries/pfas">P<strong>FAS analysis solution</strong></a> that aligns with these regulatory methods is essential for accurate detection and quantification.</p><ul><li><strong>EPA Method 1633</strong> is designed for use under the Clean Water Act (CWA) to measure the levels of 40 different PFAS in various sample types including water, soil, biosolids, sediment, and biological tissue, using <strong>LC-MS/MS</strong>. The tool was designed to address one of the main challenges in GC×GC-MS workflows: extracting meaningful chemical information from highly complex multidimensional datasets while reducing dependence on proprietary software. GcDUO combines the strengths of <strong>PARAFAC</strong> and <strong>PARAFAC2</strong> chemometric algorithms to perform automated peak detection, deconvolution, compound annotation, and quantification across large batches of samples.</p><p>The workflow begins with raw GC×GC-MS data imported from CDF files and converted into a four-dimensional matrix. Regions of interest (ROIs) are automatically selected using a watershed algorithm, allowing relevant chromatographic features to be isolated from the background. PARAFAC decomposition is then applied to separate overlapping compounds into their individual first-dimension retention times, second-dimension retention times, and mass spectra. Compound identification is performed using spectral similarity calculations, retention index information, and comparison against reference libraries, creating a consensus-based annotation strategy that improves confidence in identifications.</p><p>To further improve quantitative performance, GcDUO incorporates a constrained PARAFAC2 approach that can model chromatographic peak shifts between samples while preserving accurate peak integration. The framework was evaluated using benchmark GC×GC datasets containing fragrances, allergens, food-related compounds, and breath-analysis samples. Validation studies showed strong detection and annotation performance, including successful identification of most compounds in independent datasets such as FruityBeer and BreathMix. Quantitative comparisons with the widely used <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/products/41"><strong>ChromaTOF software</strong></a> demonstrated highly comparable retention times and peak areas for a range of volatile compounds, indicating that GcDUO can deliver results similar to established commercial platforms.</p><p>The authors conclude that GcDUO provides a powerful open-source alternative for GC×GC-MS data processing. The framework supports automated deconvolution, annotation, and quantification without requiring strict trilinearity assumptions often associated with traditional chemometric approaches. Although computational cost and user expertise remain important considerations, GcDUO offers performance comparable to gold-standard commercial software while expanding access to advanced GC×GC-MS data analysis for metabolomics, environmental, food, fragrance, and other analytical applications. The software is freely available through GitHub, supporting broader adoption and further development by the scientific community.</p><h3><strong>3. Shimadzu: Discrimination of Coffee Bean Species Based on Aroma Compounds</strong></h3><ul><li>Application note</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/an_01_01123_en_c8ac025ba9.pdf"><strong>Full PDF for download</strong></a></li></ul><h5><strong>User Benefits</strong></h5><ul><li>Qualitative analysis of aroma compounds can be easily performed using the Smart Aroma Database.</li><li>Intuitive statistical analysis and exploration of characteristic compounds are possible using eMSTAT Solution.</li><li>The AOC -6000 multifunctional autosampler system enables the analysis of aroma compounds without sample pretreatment.</li></ul><p>Common coffee bean types are broadly classified into two types: Coffea arabica (Arabica) and Coffea canephora (Robusta). Among them, Arabica is superior in flavor and is highly favored as a specialty beverage. However, in recent years, there have been multiple reports of products sold claiming to be 100% Arabica while being adulterated with Robusta<sup>1)</sup>. A reported method for discriminating between the two is the analysis of lipophilic extracts of coffee beans using a benchtop NMR spectrometer<sup>1)</sup>. This method uses 16-O-Methylcafestol, which is present in Robusta but almost undetectable in Arabica, as a marker. However, it requires complicated pretreatment such as extraction and concentration.&nbsp;</p><p>Therefore, in this Application News, we investigated a method to discriminate between Arabica and Robusta more easily and rapidly using GC-MS/MS and the statistical analysis software eMSTAT Solution.</p><h5><strong>Sample and Analytical Methods&nbsp;</strong></h5><p>Commercially available coffee beans roasted at the same roastery were used as samples. Four brands of Arabica (Kilimanjaro, Brazil Bourbon, Mandheling G1, Brazil No.2 #18) and two brands of Robusta (Vietnam Robusta, Java Robusta) were prepared (Table 1). Each brand was individually crushed with a mill, and 1 g was weighed and sealed in a screw vial.</p><p>Volatile compounds were concentrated and introduced into the GC-MS using the HS-SPME method and then analyzed under the conditions registered in the GC-MS(/MS) Smart Aroma Database (Table 2). The <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMSTQ8040+NX"><strong>GCMS-TQTM8040 NX</strong></a> and the <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=AOC-6000"><strong>AOC-6000 multifunctional autosampler</strong></a> system were used as the analytical instruments (Fig. 1). To verify whether rapid analysis is possible, a temperature ramp of 10˚C/min was adopted, and a low-polarity column was used to obtain stable retention times for each compound. Data acquisition was performed in Scan mode or MRM mode, and each sample was analyzed three times.</p><h4><strong>Exploration of Characteristic Aroma Compounds for Arabica and Robusta&nbsp;</strong></h4><p>Next, we investigated which compounds contribute to the discrimination between Arabica and Robusta. The PCA score plot (Fig. 4) reflects the differences in the tendencies of each group, with Arabica distributed on the left side and Robusta on the right side, clearly separated along the first principal component (PC1) axis. Referring to the corresponding loading plot, compounds located on the left side of the origin tend to have higher area values in Arabica, which is also positioned on the left in the score plot. On the other hand, compounds on the right side tend to show higher values in Robusta, which is located on the right in the score plot. In other words, the difference in the balance of these compoundsisthe main factorseparating the two groups. First, Arabica was commonly rich in compounds such as 5-Methyl furfural, Acetoin, and Furaneol acetate. MS chromatograms analyzed using LabSolutions Insight GCMS are shown in Fig. 7. In eMSTAT Solution, not only score plots and loading plots but also box plots can be easily drawn, making it possible to easily visualize and compare the distribution tendencies of each compound. From the box plots of the above compounds (Fig. 8), it was visually confirmed that all of them are contained in abundance in Arabica. These compounds are sometimes described as having a roasted or creamy quality, and it can be inferred that this result supports the characteristics of Arabica, which issuperior in aroma and acidity.&nbsp;</p><p>On the other hand, a compound found in abundance in Robusta was p-Vinylguaiacol. This compound is known to cause an offflavor similar to a medicinal odor in sake, and its content was low in Arabica. From this, it was suggested that p-Vinylguaiacol may be one of the compounds contributing to the bitterness and offtastes characteristic of Robusta.</p><h4><strong>Conclusion&nbsp;</strong></h4><p>In this Application News, we investigated the discrimination of coffee bean types based on aroma compounds using the triple quadrupole gas chromatograph mass spectrometer <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=GCMSTQ8040+NX"><strong>GCMSTQ8040 NX</strong></a> and the GC-MS(/MS) <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=1&amp;search=Smart+Aroma+Database"><strong>Smart Aroma Database</strong></a> for aroma analysis.&nbsp;</p><p>Four types of Arabica and two types of Robusta were analyzed as samples. Analysis using Scan data confirmed a tendency for the two types to separate. Subsequently, data was acquired in MRM mode, which offers higher sensitivity and selectivity, and detailed analysis was performed using the statistical analysis software eMSTAT Solution. As a result, the two groups were clearly separated in PCA and cluster analysis, demonstrating the effectiveness of discrimination by this method. Additionally, a discrimination model was developed and validated, confirming that highly accurate discrimination is possible.&nbsp;</p><p>From the above results, we conclude that the discrimination of coffee beans by aroma compound analysisis highly feasible using this method. Furthermore, this method enables a comprehensive workflow, from discrimination to the identification of characteristic compoundsfor each type.</p><h3><strong>4. Waters Corporation: Improvements to the Analysis of PBDEs in Environmental Matrices: Transitioning from EI Magnetic Sector to GC-APCI TQ MS/MS</strong></h3><ul><li>Poster</li><li><a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/labrulez-bucket-strapi-h3hsga3/720009372_7abfa9f0e9.pdf"><strong>Full PDF for download</strong></a></li></ul><p>This poster evaluates the transition of polybrominated diphenyl ether (PBDE) analysis in environmental samples from traditional electron ionization high-resolution magnetic sector GC-MS to a modern <strong>GC-APCI triple quadrupole MS/MS (GC-APCI TQ MS/MS)</strong> workflow. PBDE flame retardants are highly regulated due to their environmental persistence and potential health risks, making accurate and reliable analytical methods essential. The study investigated whether a GC-APCI MS/MS approach could match or exceed the performance of established GC-HRMS methods while offering greater flexibility and modernization opportunities.</p><p>The method was developed on a <a target="_blank" rel="noopener noreferrer" href="https://lcms.labrulez.com/products/554"><strong>Waters Xevo TQ Absolute mass spectrometer</strong> </a>equipped with APCI ionization and an <a target="_blank" rel="noopener noreferrer" href="https://gcms.labrulez.com/library?mainauthorItems=10&amp;search=8890"><strong>Agilent 8890 GC</strong></a>. A key innovation was the use of <strong>nitrogen as the carrier gas instead of helium</strong>, helping laboratories reduce dependence on helium supplies and mitigate increasing helium costs. By applying nitrogen flow ramping, the authors maintained chromatographic resolution and runtime comparable to the reference helium-based method without requiring changes in column dimensions. The approach also reduced the risk of thermal degradation of highly brominated compounds such as BDE 209 and achieved faster elution while preserving peak shape and separation quality.</p><p>For quantitative analysis, up to six MRM transitions per analyte were monitored, taking advantage of the characteristic isotope patterns of brominated compounds. The use of multiple transitions improved selectivity and sensitivity, while Waters waters_connect software enabled summation of MRM signals to enhance signal-to-noise ratios near detection limits. Excellent calibration performance was achieved across more than three orders of magnitude, with correlation coefficients above 0.999 for all analytes. Internal standards and recovery standards demonstrated strong reproducibility, and quality control samples showed precision typically below 15% RSD and acceptable accuracy across low, medium, and high concentration levels.</p><p>The authors conclude that an all-nitrogen GC-APCI TQ MS/MS configuration can successfully meet the performance criteria established for PBDE analysis by reference EI GC-HRMS methods. The workflow maintains critical chromatographic separations, delivers accurate quantitative results, and offers a platform that could potentially combine PBDE determination with other halogenated flame retardants in a single analytical method. These findings demonstrate that GC-APCI TQ MS/MS represents a robust and practical modernization strategy for environmental PBDE analysis.</p></div></div></div></div></div></div></section></div></div></div></div></div></div></section>

<p><strong>Non-authorized presence of ethylene oxide (ETO) and 2-chloroethanol (2CE) in sesame seeds and other food commodities is a new emerging food safety issue. First notification about the presence of ETO in sesame seeds dates from August 2020, and since then over 500 new notifications were published on the European Rapid Alert Food and Feed Safety (RASFF) portal. To ensure that foods are safe for consumption, food industry and enforcement agencies need reliable and robust methods to detect ETO and 2CE in foods preferably at levels at or below the set maximum residue limit of 0.05 mg/kg.</strong></p>
<p>For the determination of these contaminants a combination of a QuEChERS sample preparation method and a GC-MS/MS analytical method was proposed by the EU Reference Laboratories for Residues of Pesticides. However, several challenges are encountered when it comes to the analysis of ETO and 2CE in various food products. The high volatility of ETO requires the use of a dedicated column to ensure separation from possible interference from acetaldehyde. </p>
<p>Secondly, QuEChERS extracts often contain high amounts of non-volatile material which can accumulate in the inlet liner and column inlet, affecting the accuracy and robustness of the analysis. Integration of the Automated Liner Exchange (ALEX) option in the analysis of ETO and 2CE allowed to exchange the GC liner without stopping the analysis sequence and therefore with no downtime related to inlet maintenance.</p>
<p>Additionally, the analytical column was further protected by the integration of a pre-column with backflushing option. This allowed to remove high boiling compounds which reach the analytical column. Back-flushing was carried out while the analysis was occurring on the analytical column, keeping sample throughput unaffected. </p>
<p>With this dedicated set-up based on an Agilent 8890 GC, an Agilent 7010 triple quadrupole MS and a Gerstel MPS sampler, a robust analytical solution was obtained, ensuring less downtime related to inlet maintenance, column changing and/or MS source cleaning. Limits of detection in the low range of µg/kg with excellent recovery and repeatability were obtained for sesame and curcuma samples. </p>

<p>The present research is focused on the comparison between the use of hydrogen and helium carrier gases, within the context of flow-modulation fast comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry. All experiments were carried out by using the following column set: low polarity with dimensions 10 m × 0.25 mm ID × 0.25 µm df and medium polarity with dimensions 2 m × 0.10 mm ID × 0.10 µm df. Fundamental gas chromatography parameters (efficiency, resolution) were measured under different experimental conditions, using the two carrier gases. Efficiency was measured in both the first (using C12 alkane) and second (using naphthalene) dimensions; after defining the optimum carrier gas conditions, a sample of diesel was analyzed, with the overall separation evaluated to make final conclusions on the analytical performances. Finally, a comparison was made between mass spectral profiles obtained analyzing fatty acid methyl esters and pesticides.</p>

<h4><strong>Switch to Fast GC Easily - Theory and Practice</strong></h4>
<p>There a number of ways that the analysis time can be reduced. It all depends on how well the components are separated in the present application. Most easy is to trade in some resolution for speed using higher gas velocities or using a shorter column. If the exact separation must be maintained, a choice has to be made between using hydrogen as carrier gas, or use a smaller ID capillary column (or a combination). What is important is that in the new method, the same chromatogram (and peak elution order) must be obtained. Using the EZGC method translator, you can simply calculate the oven program you need for that. Also, in the ProEZGC chromatogram modeler, you can see the impact of any changed parameter, just using your laptop.</p>

For a number of reasons, including safety, cost, convenience, and analytical throughput, more and more labs are abandoning hydrogen cylinders in favor of hydrogen gas generators to supply the gases they require for their analyses.
