<p><strong>Discoveries from IOCB Prague now have a greater chance of being put into practice starting today, and patients with previously untreatable diseases have stronger hopes of receiving effective therapy. A unique translational center connecting science with its commercial applications – Pharmtheon – is now operational, marking a significant development in Czech science.</strong></p>
<p>State-of-the-art laboratories have found their home at Harfa in Prague, where preclinical research will be conducted to bring promising molecules closer to the development of important medications.</p>

<p>The transfer of technologies into practice has long been supported by the Czech government, which has identified this area as one of its priorities in its program statement. The first project to be developed at the center is the development of substances that reduce the frequency of mutations in cancer cells. These mutations are the cause of resistance to some drugs used in the chemotherapy of cancer patients. This area is highly relevant and is currently somewhat overlooked.</p>
<p><em>&quot;The development of new drugs is particularly challenging in the environment of academic institutions, where basic research is the priority. This may not align with efforts to promote the transfer of inventions into practice. Therefore, I consider the creation of a suitable environment that combines expert knowledge, financial resources, and technical equipment to be crucial. That is also the main reason why Pharmtheon was established,&quot;</em> explained Martin Fusek, the initiator of the center&#39;s establishment.</p>

<p>The creation of new drugs first requires an understanding of the mechanism of disease development and then attempts to influence these mechanisms with administered drugs. If substances that can positively influence certain pathological processes at the cellular level are found, these substances need to be optimized in terms of effectiveness, side effects, duration of action, and many other properties in preclinical research. This process is called translational research. Now, thanks to Pharmtheon offering professional support to translational research and enabling more efficient collaboration among all involved parties, even better results can be expected.</p>
<p>Pharmtheon Center is part of the IOCB Tech family, a subsidiary of the Institute of Organic Chemistry and Biochemistry of the Czech Academy of Sciences in Prague.</p>

<p>We are modern well equipped laboratory with the technical background able to perform most of the currently used <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/untargeted-proteomics.24?t=s"><strong>untargeted</strong></a> and <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/targeted-proteomics.25?t=s"><strong>targeted</strong></a> proteomic measurements. We provide full proteomics service, including protein isolation from the supplied sample to evaluation and consultation of the measured data. Since 2015, we handle approximately 300 orders per year (roughly 3000 samples). Results based on measurements from our laboratory were used in dozens of publications. <p>As a service, we provide <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/targeted-metabolomics.26?t=s"><strong>targeted</strong></a> and <a href="https://www.biocev.eu/en/services/omics-mass-spectrometry-core-facility.6/untargeted-metabolomics.30?t=s"><strong>untargeted</strong></a> analysis of small molecules. Thanks to the combination of equipment for <strong>LC-MS</strong> and <strong>GCxGC-MS</strong>, it is possible to measure a wide range of small molecules. We use several types of <strong>C18</strong> and <strong>HILIC</strong> stationary phase columns on LC. For GC, we have various columns for different types of analytes. Individual measuremnts must always be consulted in advance, an individual approach and method must be chosen for each group of molecules. <h4><strong>Untargeted proteomics</strong></h4>
<h5><strong>Up to 8000 identified proteins in one injection.</strong></h5>
<p>A basic type of proteomic experiment. Complete lysate is digested with trypsin. The resulting peptides are desalted and separated on a reverse phase column. Due to the low amount of proteins and the high complexity of the sample, liquid chromatography is currently most often used in nanoflow - hundreds of nanoliters per minute. Liquid chromatography is directly connected to the electrospray of the Orbitrap Fusion or Orbitrap Ascend mass spectrometer. The mass spectrometer measures alternately in two modes: MS¹, where peptide masses are measured in the Orbitrap with high resolution and accuracy, and MS², in which peptides selected from the previous MS¹ scan are fragmented. These are measured either again in the Orbitrap or in the ion trap. The ion trap provides lower accuracy and resolution than the Orbitrap, but is faster and more sensitive.</p>

<h4><strong>Targeted proteomics</strong></h4>
<h5><strong>Accurate quantification of selected proteins.</strong></h5>
<p>Method aims to validate the results from an untargeted approach. It is performed on a triple quadrupole in SRM mode or using the MRM method on an Orbitrap. It is an an alternative approach to common techniques such as western blot. The advantage of this approach is the possibility to analyze several proteins (dozens) at once during one injection. This eliminates the problem of finding suitable antibodies.</p>

<p><strong>The targeted proteomics service is always project specific and we offer it after prior meeting with the customer.</strong></p>
</blockquote>
<p>Parallel analysis of peptides labeled with heavy amino acids is essential. These serve as an internal standards.</p>
<p>The amount of proteins that can be targeted in one injection using a triple quadrupole, is strongly dependent on the concentration of the target proteins in the sample. The lower the expression of the target proteins, the less it is possible to analyze them in one injection due to the need for higher loading times. Information of fragmentation properties of targeted peptides is also important. This is usually obtained from an untargeted experiment, which should precede the targeted proteomic analysis. If the protein of interest was not detected during an untargeted experiment, then its targeted analysis is very difficult. However, in some cases fragmentation information can be obtained from publicly available databases.</p>

<h4><strong>Untargeted metabolomics</strong></h4>
<p>The goal of untargeted metabolomics is to cover the widest possible spectrum of metabolites present in the sample, their identification, and quantification. The development of this approach was made possible by the recent advances in instrumentation and computing. Our laboratory equipment enables non-targeted analyses by liquid and gas chromatography.</p>

<p><strong>We offer untargeted analyses of metabolites in various biological matrices and basic statistical evaluation of the data (Figure 2, 3, 4).</strong></p>

<p>HPLC-MS/MS (Figure 2): Analyzes are performed on Thermo Scientific equipment on an Ultimate 3000 liquid chromatograph coupled to an Orbitrap Fusion mass detector. The measured data are evaluated by the Compound Discoverer software.</p>
<p>GCxGC-MS (Figure 3): Analyzes are performed on a two-dimensional comprehensive chromatograph connected to a Pegasus 4D mass detector. We evaluate the data in ChromaTOF v4.5 software.</p>
<p>The liquid chromatography method covers non-volatile substances, generally polar, soluble in water, and polar organic solvents (methanol, acetonitrile...). The gas chromatography method is more suitable for analyzing stable volatile non-polar substances up to a weight of approx. 1000 Da. The range of measured substances can be increased for both methods eg., by chemical modifications (derivatization) of the analyzed molecules, which adjust their polarity and volatility (for GC) and/or stability (for LC). We choose a suitable technique or a combination of several techniques based on prior consultation with the customer.</p>
<p>The untargeted analysis results in a list of substances found in the samples, a proposal of probable structures, and an indication of the quantity of individual compounds. We also provide a basic statistical evaluation focusing on looking for differences between the tested sample groups (Figure 4).</p>

<h4><strong>Targeted metabolomics</strong></h4>
<p>Targeted metabolomics includes analyses of selected metabolites, which are then detected selectively and with greater sensitivity than applying an untargeted approach. It is also possible to quantify substances both relatively and absolutely. In the case of absolute quantification, it is necessary to supply a suitable labeled analog of the determined substance.</p>

We can develop a method for any substance or group of substances capable of being detected by liquid and gas chromatography techniques (both connected to a mass detector or a UV detector in liquid chromatography). In the case of the development of a new method, it is necessary to supply the standards of these substances.</p>

<p>Mass spectrometer combining Quadrupole, Ion trap and Orbitrap. The quadrupole is used as a mass filter for the selection of precursors, after which the instrument branches and the ions can travel either to the Orbitrap - a very accurate and high-resolution mass analyzer that uses the Fourier transform to determine their mass, or to the collision cell, where they can be fragmented. From there, the path leads either back to the Orbitrap or further to the linear ion trap. This serves as the second mass analyzer in the device. It is less accurate and low-resolution, but provides greater sensitivity and the possibility of ion fragmentation. Fragmentation products can be isolated in the trap and fragmented again. This enables analysis in MS³ and even higher levels of fragmentation.</p>

Introducing BIOCEV OMICS Mass Spectrometry Core Facility

<p><strong>Environmental analytical laboratories play a key role in protecting the environment, and it is therefore essential that they themselves take action towards sustainability. This EnviroMail focuses on the annual plastic savings that one unit of European laboratories have achieved through innovative approaches and changes in working practices. Specific measures such as replacing single-use plastics with reusable alternatives, optimizing processes and using modern technologies were used for plastic saving. Our achievements show that SUSTAINABILITY is not just a word for us, but we are continuously working on a greener future.&nbsp;</strong></p><p>In recent decades, plastic has become an integral part of our lives. Its practical properties such as low weight, durability, and low cost have led to its global mass use. However, the increase in plastic waste, especially single-use plastics, has caused significant environmental problems. In the laboratory, we also use a lot of laboratory utensils and equipment made of plastic. Most of them end up as hazardous waste after the use. <strong>Reducing the amount of plastic consumables </strong>used has become one of our company's key sustainability issues.</p><h3><strong>Problems with plastics</strong></h3><p>Plastics, particularly single-use items like plastic bottles, bags, packaging, and straws, are a major source of waste that pollutes our oceans, rivers, and land. This waste is highly resistant to decomposition, leading plastic items to persist in the environment for decades or even hundreds of years. Microplastics, tiny pieces of plastic, are not only found in the natural world but also in the food chain, posing a risk to both human and animal health.</p><h3><strong>Sustainable approaches to plastics saving</strong></h3><p>In the fight against plastic waste, there are a number of initiatives aimed at saving plastic and promoting sustainable development. Key approaches include:</p><ol><li><p><strong>Reducing plastic consumption</strong></p><p>Reducing the consumption of plastics, especially single-use plastics, is a key part of the solution to the plastic problem. This includes reducing the use of plastic consumables, or at least using smaller sizes of different containers and tubes that are then lighter in weight or substituting other materials.</p></li><li><p><strong>Plastic recycling</strong></p><p>Plastic recycling is another key tool in the fight against the plastic waste. Improving recycling processes, introducing new technologies and promoting recycled materials in industry can help to significantly reduce plastic waste. In addition, recycling plastics saves natural resources, reduces CO2 emissions and helps to save energy.</p></li><li><p><strong>Alternatives to plastics</strong></p><p>The development of alternative materials to plastics is a further step towards sustainability. For example, bioplastics, made from renewable raw materials such as corn or potatoes, are a greener alternative to traditional plastics. Some companies are also focusing on creating packaging that is compostable or easily recyclable.</p></li></ol><h3><strong>Towards ALS Net Zero 2050</strong></h3><p>Saving plastics is a key to achieving the Sustainable Development Goals. In one of the main European laboratories, it was managed to reduce plastic waste production by <strong>1,758 kg </strong>in the last financial year 2023/2024. When the weight of plastic is taken into consideration, the saving in amount of waste is significant.</p><h3><strong>Plastics saving areas:</strong></h3><ul><li>Use of smaller disposable plastic test tubes.<br><strong>Total savings: 915 kg</strong></li><li>Replacement of plastic containers with containers made of bioplastic.<br><strong>Total savings: 412 kg</strong></li><li>Ensuring recycling of used plastic products.<br><strong>Total savings: 152 kg</strong></li><li>Replacing plastic packaging with paper packaging that is more easily recycled.<br><strong>Total savings: 145 kg</strong></li><li>Reducing the consumption of plastic containers.<br><strong>Total savings: 58 kg</strong></li><li>Purchase of non-packaged laboratory syringes for water sample filtration.<br><strong>Total savings: 47 kg</strong></li><li>Discontinuation of the purchase of bottled water.<br><strong>Total savings: 20 kg</strong></li><li>Implementation of electronic documentation management system, reduction of EURO packaging consumption.<br><strong>Total savings: 10 kg</strong></li></ul><h3><strong>Conclusions</strong></h3><p>Saving plastics is an essential step towards protecting the environment and promoting sustainable development. It is a challenge that requires a coordinated effort by all. Reducing plastic consumption, improving recycling processes, promoting alternative materials and changing consumption habits are key factors in ensuring a sustainable future for our planet. By continuing to work towards these goals, we can help to protect natural resources, improve health and ensure a good quality of life for future generations.</p>

<p><strong>Virtually no laboratory today can do without a source of purified water. The amount produced and the required purity depend on a number of factors, but it is always true that we expect stable performance and reliable function from water treatment equipment.</strong></p><p>While distillation apparatuses used to dominate, today a combination of flow purification techniques is usually used to prepare purified water for laboratories, the specific list of which depends primarily on the required final water purity. These include filtration, adsorption, removal of dissolved substances by reverse osmosis, deionization (using ion exchangers or electrochemically), photooxidation, sterilization by UV radiation and, last but not least, ultrafiltration.</p><h4><a target="_blank" rel="noopener noreferrer" href="https://watrex.com/kategorie-produktu/water-purification/?utm_source=labrulez&amp;utm_medium=propagace&amp;utm_campaign=labrulez_clanky"><strong>Overview of Watrex water systems</strong></a></h4><p>In addition, modern devices often allow continuous monitoring of water quality in real time, which significantly increases control over the process. However, a number of complications can still occur - and that is why RNDr. Roman Kysilka from <a target="_blank" rel="noopener noreferrer" href="https://watrex.com/?utm_source=labrulez&amp;utm_medium=propagace&amp;utm_campaign=labrulez_clanky"><strong>Watrex Praha</strong></a> provides an overview of the basic principles of proper operation and advice on how to recognize and solve the most common<br>problems (Troubleshooting).</p><h4><strong>How to operate a water treatment plant in a laboratory without problems</strong></h4><p>There are certain common principles that apply to the use of all equipment for the preparation of purified water. The most important ones are summarized in the following points:</p><ol><li>Recommendations before purchasing equipment - choose a model with a slight power reserve that produces water of the quality you need. The purer the water, the higher the operating costs. Use water purified by special procedures only where necessary.</li><li>Develop a maintenance plan for your equipment and its documentation. Designate a person responsible for the operation of the equipment. For low-use and basic equipment (e.g. reverse osmosis systems), annual periodic maintenance is sufficient, while for critical applications, daily water quality checks may be necessary. Establish an operating logbook, or at least record the checks and maintenance on a label placed on the equipment.</li><li>All models of devices for preparing purified water from tap water contain filters that remove suspended solids. Depending on the degree of contamination of the water from the water supply and the volume of treated water, the replacement period varies considerably. The replacement period for a specific installation is usually recommended by the supplier, but it is advisable to perform a simple visual inspection. A contaminated filter is easy to identify.</li><li>If a reverse osmosis (RO) membrane is used in the device, attention should be paid to its proper functioning. The service life of the RO membrane is several years with proper maintenance of the protective filters. If the device is not equipped with a reverse osmosis process efficiency measurement, it is advisable to check the performance, efficiency and yield of this purification stage at least once a year.</li><li>To achieve a low content of dissociated compounds (ions), filters filled with an equimolar mixture of cation and anion are most often used. More modern devices alternatively use electrodeionization. If the device includes a deionization stage, it should always be equipped with a conductivity measurement of the produced water, or at least an indication of exceeding the limit value of specific conductivity. In daily use, it is advisable to check the conductivity at least once a day, in other cases at each first sampling.</li><li>Most devices for the preparation of purified water are not sterile and are populated to varying degrees by microorganisms, which is usually accompanied by a higher value of organically bound carbon (TOC) in the water. This may not be a problem for basic laboratory applications, but if we require water that is sufficiently pure for HPLC, work with tissue cultures, molecular genetic experiments, and the like, the device must include technology to minimize the impact of organisms. This includes the inclusion of UV radiation sterilization sources, the application of ozone generated by UV emitters at 184 nm, microfiltration and ultrafiltration. For proper function, it is important to adhere to the component replacement interval recommended by the device manufacturer (either in terms of time or according to the volume of treated water) for all these techniques. Otherwise, a malfunctioning purification stage is often discovered only on the basis of false experiment results.</li><li>All types of equipment should be periodically (once a year, once every two years - depending on the intensity of use and the potential risk of endangering the experiments) verified whether they meet the parameters expected of them. For systems with their own measurement, it is necessary to check whether the measurement results of the built-in sensors provide valid values.</li></ol><p><img src="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/Watrex_8_obr_2_88c7487928.jpeg" alt="Watrex: Fig. 2: Illustrative image of possible consumables used in some water systems." srcset="https://storage.googleapis.com/labrulez-bucket-strapi-h3hsga3/thumbnail_Watrex_8_obr_2_88c7487928.jpeg 234w," sizes="100vw" width="1600" height="1068"></p><h4><strong>Troubleshooting</strong></h4><p>Even with careful maintenance and following operating recommendations, there may be times when your water treatment equipment does not function as expected or shows signs of a problem. Below are tips for identifying and resolving some common problems. Before you begin any work on your equipment, make sure you understand its function and, if necessary, consult the manual provided by the manufacturer (perhaps as a first step).</p><h5><strong>1. Basic Check</strong></h5><p>Before you start looking for more complex causes, perform the following basic checks:</p><ul><li><strong>Power and Water Supply</strong>: Check that the equipment is properly connected to the electrical network and that it is actually receiving electricity. Verify that the main water inlet valve is open and that the inlet water pressure meets the requirements of the equipment.</li><li><strong>Visual Check</strong>: Inspect the equipment for visible water leaks, loose or damaged hoses and connections. Check that filter covers or other parts are not visibly damaged.</li><li><strong>Indicators on the device</strong>: Pay attention to any status lights, displays, or alarms. Many devices indicate specific problems (e.g., filter replacement required, low pressure, poor water quality) using codes or symbols. Refer to the manual for the meaning of these indications.</li><li><strong>Logbook/Maintenance Records</strong>: Refer to the logbook or maintenance label. When was the last maintenance performed? Have any components been changed recently? This information may help identify the cause of the problem.</li></ul><h5><strong>2. Specific Problems and Possible Solutions</strong></h5><ul><li><strong>Low or no flow of purified water:</strong><ul><li><i>Possible cause</i>: Clogged pre-filter (mechanical filter). Low inlet water pressure. Closed valve in the system. Pump problem (if included in the system). Heavily clogged or damaged RO membrane. Clogged final (sterile) filter.</li><li><i>Possible solution</i>: Check and replace pre-filter (visual inspection as mentioned above). Verify inlet water pressure. Go through all valves and make sure they are in the correct position. Verify pump function according to manual. Check RO membrane performance (if possible, see previous text) – may require replacement. Replace final filter.</li></ul></li><li><strong>Insufficient water quality (High conductivity):</strong><ul><li><i>Possible cause</i>: Deionization cartridge (ion exchanger) capacity exhausted. Reduced RO membrane efficiency (insufficient ion removal). Problem with electrodeionization (EDI) module. Conductivity measuring sensor failure or error.</li><li><i>Possible solution</i>: Check conductivity value on display or indicator (as mentioned in previous text). If value is out of limit, replace deionization cartridge(s). Verify RO stage efficiency (by measuring conductivity before and after RO membrane, if system allows). Verify EDI module function according to manual (may require service). Consider checking/calibrating measuring sensor (usually performed by service technician).</li></ul></li><li><strong>Increased TOC (Total Organic Carbon) or suspected microbial contamination:</strong><ul><li><i>Possible cause</i>: Inoperative or old UV lamp (184 nm for TOC oxidation, 254 nm for sterilization). Exhausted or damaged final filter (ultrafilter, microfilter). Biofilm formation in the system, piping or tank (if used). High levels of organics or microorganisms in the inlet water.</li><li><i>Possible solution</i>: Check the operating hours or installation date of the UV lamp and replace it if it has exceeded the recommended life (according to the maintenance schedule). Replace the final filter. Sanitize the system according to the manufacturer’s recommendations (may include chemical cleaning or hot water flushing). Consider installing additional inlet water pretreatment if it is consistently poor.</li></ul></li><li><strong>Water leaks from the system:</strong><ul><li><i>Possible cause</i>: Loose fittings or hose connections. Damaged seal (e.g. at the filter housing). Cracked filter housing or other component (e.g. due to frost or mechanical damage). Too high water pressure in the system.</li><li><i>Possible solution</i>: Immediately shut off the water supply and disconnect the appliance from the electricity supply. Carefully locate the leak. Carefully tighten the relevant fitting or connection (do not overtighten, as this may damage the thread). If the seal or component is damaged, it must be replaced with an original spare part. Check the inlet water pressure and the function of the pressure reducing valve (if installed).</li></ul></li><li><strong>The appliance reports an alarm or warning:</strong><ul><li><i>Possible cause</i>: The system has detected a specific operating deviation or the need for maintenance.</li><li><i>Possible solution</i>: Always follow the instructions in the manufacturer’s manual for the specific alarm code or symbol. This may indicate the need to replace consumables (filter, cartridge, UV lamp), low/high pressure warning, poor water quality, internal error or the need to perform sanitation. If the steps in the manual do not resolve the problem, contact the service.</li></ul></li></ul><h5><strong>3. When to Contact Service Support</strong></h5><p>Although many common problems can be solved by yourself based on this manual and the device manual, in some cases it is necessary to contact an authorized service center or supplier:</p><ul><li>If the basic troubleshooting steps have failed.</li><li>If the problem recurs even after performing the recommended steps.</li><li>If any major water leakage that cannot be easily stopped and repaired.</li><li>If the device reports serious error codes that are not described in the user manual.</li><li>If it is necessary to replace key components such as the RO membrane, EDI module, pump, or main control unit.</li><li>For regular validation and calibration of measuring sensors (conductivity, resistance, TOC), as mentioned in the previous text.</li><li>If you are unsure of the cause of the problem or the correct repair procedure.
