How to link a calibration to a chromatogram in Clarity Chromatography Software

Proper calibration management is essential for generating accurate quantitative results in chromatographic analysis. Clarity Chromatography Software allows users to assign calibrations either to chromatograms that have already been measured or to methods used for future acquisitions. This article provides a detailed, step-by-step guide to linking calibrations within Clarity, ensuring immediate and reliable calculation of analytical results.

1. Overview of Calibration Linking in Clarity

Calibration represents the mathematical relationship between detector signal and analyte concentration. Correctly linking calibration ensures that:

  • Quantitative results are calculated automatically.
  • Chromatograms are processed using the intended calibration curve.
  • Standard injections can update or supplement existing calibration levels.

Clarity supports two primary workflows:

  1. Linking calibration to an already measured chromatogram.
  2. Linking calibration to a method, which then applies automatically to all future chromatograms measured with that method.

2. Linking Calibration to an Already Measured Chromatogram

2.1 Accessing the Chromatogram Window
  1. Open the chromatogram you wish to process.
  2. Switch to the Results tab.
2.2 Assigning Calibration
  1. In the section labeled Common for All Signals, click Set.
  2. A calibration selection dialog opens.
  3. Choose the desired calibration file or calibration set.
2.3 Result Calculation

Once the calibration is linked:

  • Clarity immediately recalculates results.
  • Correct peak quantitation becomes visible in the Results table.
  • No additional processing steps are required — provided that the calibration itself is valid and properly configured.

3. Linking Calibration to Newly Measured Chromatograms via the Method

3.1 Opening the Method Setup
  1. Navigate to Method Setup.
  2. Open the method you plan to use for measurement.
3.2 Linking Calibration in the Method
  1. Go to the Calculation tab.
  2. Click Set.
  3. Select the intended calibration.
3.3 Saving and Sending the Method to the Instrument
  1. Save the modified method.
  2. Send the method to the connected instrument.
  3. The method can now be used in both:
    • Single Analysis
    • Sequence Acquisition

4. Important Settings During Measurement

4.1 Sample Type Behavior

When starting a new measurement, pay close attention to the Sample Type setting:

Unknown

  • Calibration is linked only.
  • No change is made to the calibration file.

Standard

  • The specified calibration level will be recalibrated using this chromatogram.
  • Useful for:
    • Adding new levels,
    • Updating existing calibration curves,
    • Maintaining calibration accuracy.
4.2 Starting the Measurement
  1. Confirm the method settings.
  2. Select Single Analysis or Sequence.
  3. Start acquisition.

5. Reviewing Newly Measured Chromatograms

After measurement completes:

  1. Open the newly measured chromatogram.
  2. The previously selected calibration is automatically linked.
  3. If the calibration is valid, results are calculated immediately and automatically.

This automation ensures consistency across data sets and minimizes user error.
